摘要
目的为了弥补微弧氧化缺陷以及提高其生物活性,在镁超声微弧氧化(UMAO)表面制备一层植酸掺杂葛根素粒子的植酸复合涂层。方法 UMAO,植酸(phytic acid,PA)作为对照组,PA-10μg/mL葛根素为实验组,通过带能谱SEM观察表面涂层的形貌以及进行元素分析,以小鼠成骨细胞(MC3T3-E1)为实验细胞,并通过CCK-8试剂盒检测试件表面细胞增殖和黏附能力,并采用激光共聚焦显微镜观察成骨细胞在凃层表面的铺展情况。结果在黏附和增殖实验中,PA涂层的吸光度大于MAO涂层,PA-10μg/mL涂层吸光度值均大于对照组,且实验结果具有统计学意义。相对于MAO涂层,PA涂层,细胞在PA-10μg/mL涂层上充分铺展,细胞核数量最多。结论纯镁超声微弧氧化植酸-10μg/mL葛根素复合膜层的生物活性提高了,具有最佳的生物相容性。
Objective In order to compensate for the defects of MAO coating and improve its biological activity, a composite coating of phytic acid with puerarin was prepared on the surface of magnesium by ul- trasonic micro-arc oxidation. Methods The UMAO and phytic acid coatings were used as a control group while PA-10 μg/mL coating is used as an experimental group. The surface morphology and elemen- tal component of coatings were analyzed by scanning electron microscope with an energy spectrum. The cell proliferation and adhesion of coatings were tested by CCK-8 using MC3T3-E1 as experimental ceils. The cytoskeleton of coatings was observed by laser confocal microscopy. Results The absorbance of the PA coating was greater than the MAO coating in the adhesion and proliferation experiments. The absor- bance of PA-10 μg/mL coating was larger than all control groups. All the experimental results were statis- tically significant. Compared with MAO coating and PA coating, cells spread fully on the PA-10 Ixg/mL coating with the largest number of nuclei. Conclusion The biological activity of PA- 10 g/mL puerarin coating of ultrasonic microarc oxidation is improved and has great biocompatibility.
