摘要
目的探讨不同浓度的葛根素溶液对纯镁经超声微弧氧化植酸处理后表面生长的成骨细胞的影响,确定最适宜成骨细胞生长的浓度。方法取四组不同浓度组:A组0 mol/L葛根素溶液,B组1×10^(-8)mol/L葛根素溶液,C组1×10^(-6)mol/L葛根素溶液,D组1×10^(-4)mol/L葛根素溶液,通过CCK-8法检测不同时间点纯镁试件表面生长的成骨细胞的黏附、增殖状况,激光共聚焦显微镜下观察细胞的早期骨架形态。结果纯镁经超声微弧氧化后植酸处理达到封孔效果,成骨细胞附着于试件生长后加入相应浓度的葛根素溶液,四组实验CCK-8黏附、增殖数据显示C组>B组>A组,A组与D组之间未见统计学差异,激光共聚焦显微镜下显示C组的细胞形态效果最好。结论 1×10^(-6)mol/L葛根素溶液为促进纯镁UMAO植酸涂层表面的成骨细胞生长的最适宜浓度。
Objective Through the comparison of different concentrations of Puerarin solution, observing the effect of growth of osteoblasts on pure magnesium ultrasonic micro arc oxidation phytic acid coating, to determine the most suitable concentration of osteoblasts growth. Method The experimental subjects were divided into four concentration groups: The group A was treated with 0mol/L puerarin, the group B was treated with 1 ×10^-8 mol/Lpuerarin, the group C was treated with 1 ×10^-6 mol/Lpuerarin, the group D was treated with 1 ×10^-4mol/Lpuerarin. CCK- 8 method was used to detect the adhesion and proliferation of osteoblasts on the surface of pure magnesium specimens at different time points, and laser confocal microscopy was used to detect cell morphology and cytoskeleon. Results Pure magnesium ultrasonic micro arc oxidized phytic acid treatment can achieve sealing effect, adding correspond concen- tration of puerarin solution after osteoblasts attaching to the specimen, CCK - 8 test showed that the OD value of group A, B and C were increased and in the turn at C 〉 B 〉 A, no statistical difference between group A and group D, and laser confocal microscopy showed cell growth status: group C was the best Conclusion The group C was optimum concentration to promote osteoblasts growth of pure magnesium ultrasonic micro arc oxidation phytic acid coating
