摘要
目的探讨红色诺卡菌细胞壁骨架(N-CWS)对小鼠皮肤创伤愈合的影响。方法建立小鼠背部皮肤创伤模型,观察小鼠伤口愈合时间和愈合率,HE染色观察创面组织学的变化,免疫组织化学染色法检测血管内皮生长因子(VEGF)和血小板源性生长因子(PDGF)的变化。结果与空白组比较,N-CWS能明显提高小鼠创伤愈合速度和愈合率(P<0.05或P<0.01)。HE染色显示N-CWS组毛细血管和胶原纤维增生明显,表皮再生完全。免疫组织化学染色显示,5d时N-CWS组VEGF表达水平明显高于空白组(P<0.05),与对照组差异无统计学意义(P>0.05);7d时N-CWS组PDGF表达水平明显高于空白组(P<0.05),低于对照组的表达水平。结论 N-CWS在创伤修复愈合过程中起明显促进作用,可能与适度调节VEGF和PDGF生长因子表达有关。
Objective To study the effects ofNocardia rubra cell wall skeleton (N-CWS) on wound healing of skin in mouse. Methods The mouse skin wound model was established. The mouse wound healing time and healing rate were recorded after the treatment with N-CWS. The change of wound histology was observed by the hematoxylin- eosin (HE) staining. The change of vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) were detected by immunohistochemical staining respectively. Results N-CWS could obviously improve the healing speed and the healing rate of wound in mouse compared with the blank group (P〈0.05 or P〈0.01). HE staining revealed that remarkable capillary and collagen fiber hyperplasia, and complete epidermal regeneration were observed in the N-CWS group. Immunohistochemical staining revealed that the expression level of VEGF in the N-CWS group was higher than that of the blank group in 5d (P〈0.05), but there was no significant difference compared with the control group. The expression level of PDGF was greatly higher than that of the blank group and lower than that of the control group in 7d (P〈0.05). Conclusion N-CWS played a significant promotion role on wound healing, it might be related to the proper promotion of the expression of VEGF and PDGE.
