摘要
本研究采用O型口蹄疫灭活病毒配合枯草芽孢杆菌喷鼻气雾免疫健康牛,并设单独使用枯草芽孢杆菌组、灭活病毒、常规疫苗免疫组和空白对照组,评价O型口蹄疫灭活病毒配合免疫增强剂枯草芽孢杆菌通过喷鼻气雾免疫健康牛对其呼吸道先天免疫力的影响。通过免疫组织化学染色显示,免疫后灭活病毒配合免疫增强剂试验组牛鼻黏膜与肺内支气管黏膜中TLR7的表达和CD3^+淋巴细胞呈极显著增多(p<0.01);而常规口蹄疫灭活苗对其TLR7的表达和CD3^+淋巴细胞数量的影响不显著(p>0.05)。ELISA检测结果显示,与对照组相比,灭活病毒配合免疫增强剂试验组牛呼吸道组织中IL-12水平呈极显著增加(p<0.01),IL-4和IL-10水平无明显差异(p>0.05);而常规口蹄疫灭活苗对牛呼吸道组织中IL-4、IL-10和IL-12的影响不明显(p>0.05)。表明O型口蹄疫灭活病毒配合枯草芽孢杆菌能够提高牛呼吸道先天免疫力。本研究为研发口蹄疫灭活病毒的鼻腔疫苗提供重要的试验数据和参考。
To explore the influence of innate immunity on cattle respiratory tract after intranasal aerosol immunized with inactivated foot-and-mouth disease virus (FMDV) along with Bacillus subtilis. In this study, we used inactivated FMDV along with B.subtilis to immune cattle by intranasal aerosol, and set up control group including single use of B.subtilis, inactivated FMDV,conventional inactivated FMDV vaccine and negative control. The results of immunohistochemistry assay showed that expressing TLR7 and CD3+ lymphocytes were significantly increased in cattle nasal mucosae and pulmonary bronchial (p〈0.01) after intranasal immunized with inactivated FMDV along with B.subtilis, but there was no significant influences (p〉0.05) in the cattle only immunized with conventional inactivated FMDV vaccine compared with other control group. At the same time, the results of ELISA showed that the level of IL-12 was significantly increased (p〈0.01, p〈0.05) and the level of IL-4 and IL-10 were not significant (p〉0.05) in cattle respiratory tract tissues after intranasal immunized with inactivated FMDV along with B.subtilis, included nasal, tracheal, pulmonary and pulmonary bronchial. However, the level of IL-4, IL-10 and IL-12 were not significant (p〉0.05) in cattle respiratory tract tissues after the inactivated FMDV vaccine immunization. These results indicated that innate immunity of the cattle respiratory tract were improved by intranasal immunization of inactivated FMDV along with B.subtilis. Our study provided important experimental data and reference for the development of the inactivated FMDV vaccine via intranasal aerosol immunization.
