摘要 【目的】本研究旨在通过DNA条形码技术,以matK基因为条形码编码序列对枸杞属种质资源10份试验材料进行鉴定分析,从而在分子水平上获得鉴定枸杞属植物的理论依据。【方法】首先利用ClustalX软件比对序列,然后用Mega7.0获得序列信息并比较序列间的差异,最后基于K2P模型构建系统发育树。【结果】matK序列总长为936 bp,保守位点933个,变异位点3个,GC含量均值为33.3%,碱基转换颠换值为1.8,其系统发育树分成了两大支,黑果、黄果变和昌吉枸杞聚为一大支,其余品种聚为一大支,各分支内部均具有较高的自展支持率。【结论】因此,matK序列可以作为初步鉴定枸杞属植物的DNA条形码。 [Objective] This study aims to analyze matK encoding gene in 10 Lycium L. plants by the technique of DNA barcoding. To do this research, the molecular evidence of identifying Lycium L. plants could be obtained. [Method] First, alignment DNA sequence by ClustalX software was used. After that, MEGA 7.0 was used to compare the differences of 10 sequences. At last, based on the model of Kimura 2-parameter to build Maximum Parsimony tree (MP tree). [Result] Total length of matK sequence was 936 bp, which included 933 conserved sites and 3 variable sites. The mean value of GC content was 33.3% and the R value was 1.8. The MP tree divideed 10 test materials into two parts. Black, yellow and Changji Lycium L. plants were classfied as one branch. Others were classified as one branch. Also, the favorability rating of bootstrap in two parts was high. [Conclusion] Therefore, matK sequence could identification Lycium L. plants approximately as DNA barcoding.
