摘要
目的:观察针刺协同抗癫痫西药(AEDs)对难治性癫痫大鼠动物行为学及P-糖蛋白(P-gp)和MDR1表达的影响。方法:难治性癫痫大鼠模型采用腹腔注射氯化锂-盐酸匹罗卡品化学点燃方法。按随机化原则将大鼠分为假手术组、模型组、西药组、针刺组、针药协同组5组,每组10只大鼠,观察周期为3个月。视频录像监测动物行为学;以Western Blot及Real-time PCR法检测治疗3个月后海马P-gp表达及MDR1 mRNA水平。结果:与模型组比较,针刺组、西药组及针药协同组大鼠癫痫发作总次数及总持续时间均显著降低(P<0.05,P<0.01),其中针药协同组下降最显著(P<0.01)。针刺组及针药协同组大鼠P-pg表达显著低于模型组(P<0.05,P<0.01),针药协同组大鼠MDR1 mRNA转录水平较模型组及西药组显著下降(P<0.05)。结论:针药协同可能改善难治性癫痫,痫四针具有增敏效应,可能与抑制耐药基因及其蛋白高表达有关。
Objective: To investigate the effects of acupuncture combined with AEDs on expression of P-gp and MDR1 in intractable epilepsy model. Methods: The rats models, which were made by intraperitioneal injection of Aluminium Choloride and Pilocapine, were randomly divided into 5 groups: sharm-operation group, model group, western medicine group, acupuncture group and acupuncture combined with AEDs group, 10 in each group. The duration of trials was 3 months, 10 in each group. The behavior of rats were continuously recorded by digital camera. The expression of P-gp and MDR1 mRNA in hippocampus were determined by Western Blot and Real-time PCR after treating for 3 months. Results: Compared with the model group, the total number and time of seizures in both western medicine group, acupuncture group, and acupuncture combined with AEDs group were significantly reduced(P0.05, P0.01). Among them, the acupuncture combined with AEDs group decreased most significantly(P0.01). The expression of P-pg in acupuncture group and acupuncture combined with AEDs group were significantly lower than that in model group(P0.05, P0.01). The transcriptional of MDR1 mRNA in acupuncture combined with AEDs group were significantly lower than that in model group and western medicine group(P0.05). Conclusion: It is suggested that acupuncture and AEDs might improve the clinical efficacy of intractable epilepsy, and Xian Sizhen has sensitization effect, which might be related to the inhibition of drug resistance gene and overexpression of protein.
