摘要
目的近年来,维吾尔族乳腺癌发病率逐年增加,且不同地域、种族、人群雌激素α受体(estrogen receptor α,ERa)基因多态性与乳腺癌易感性存在差异。本研究旨在探讨中国新疆地区维吾尔族、汉族人群ERa基因PvuⅡ及XbaⅠ位点多态性与乳腺癌风险相关性。方法选取2013-01-01—2014-09—30新疆医科大学附属肿瘤医院初治原发性乳腺癌患者200例,所有患者均经2名以上病理专家确诊,其中维吾尔族、汉族各100例。选取同期收治的年龄等一般资料相匹配的维吾尔族、汉族健康体检者各100名,应用聚合酶链反应一限制性内切酶片段长度多态性(polymerase chain reaction—based restriction fragment length polymorphism,PCR—RFLP)方法检测ERa基因PvuⅡ及XbaⅠ位点多态性分布;采用非条件Logistic回归分析PvuⅡ及XbaⅠ位点多态性与罹患乳腺癌危险性的关系。结果维吾尔族及汉族人群中,相对于XX基因型,Xx、XX、Xx/XX基因型发生乳腺癌风险显著增加,维吾尔族OR值分别为4.885(95%CI为2.626~9.008,P〈0.05)、23.63(95%CI为2.838~196.7,P〈0.05)和5.392(95%CI为2.921~9.952,P〈0.05),汉族分别为4.661(95%CI为2.511~8.468,P〈0.05)、3.608(95%CI为0.809~16.04,P〉0.05)和4.519(95%CI为2.494~8.190,P〈0.05),未见PvuⅡ位点多态性与乳腺癌风险相关性。结论ERa基因Xba Ⅰ位点多态性影响维吾尔族、汉族女性乳腺癌发生风险,Xx、XX、X基因携带者(Xx/XX基因型)乳腺癌风险增加;PvuⅡ位点多态性与乳腺癌风险无显著相关性。
OBJECTIVE Breast cancer's incidence rate in Uygur papulation has increased year by year,and association of ERα gene polymorphisms with susceptibility of breast cancer have been described with conflicting results among different regions,races and populations. We conducted this study to evaluate the association between Pvu Ⅱ (rs2234693) and Xba Ⅰ (rs9340799) polymorphisms in intron 1 of ERa gene and susceptibility of breast cancer in patients of Uyghur and Han population in Xinjiang. METHODS Breast cancer patients and health control individuals matched with the patients of Uyghur and Han papulation each 100 cases were respectively enrolled to determine Pvu Ⅰ (rs2234693) and Xba Ⅰ (rs9340799) polymorphisms by PCR-RFLP,and the association of polymorphisms with susceptibility of breast cancer was further analyzed by unconditioned logistic regression. RESULTS Increased risk to breast cancer was shown in carriers with Xx, XX, Xx/XX genotype compared with xx genotype in both Uyghur and Han papulation group, the OR values were 4.885(95%CI:2.626-9.008, P〈0.05), 23.63(95% CI:2.838-196.7, P〈0.05), 5.392(95% CI:2.921-9.952, P〈 0.05) in Uyghur papulation and 4. 661(95%CI:2. 511-8. 468,P〈0. 05),3. 608(95%CI:0. 809-16. 04,P〈0. 05), 4. 519(95%CI:2. 494-8. 190,P〈0.05) in Han papulation respectively. No association of Pvu Ⅱ polymorphisms with susceptibility of breast cancer was found. CONCLUSIONS Our data shows that ERa gene Xba Ⅰ polymorphism may influence susceptibility for breast cancer in Uyghur and Han population with increased risk to breast cancer in carriers with Xx, XX, Xx/XX genotype. Moreover,we found no association of Pvu Ⅱ polymorphisms with susceptibility of breast cancer.
