摘要
目的:研究蜘蛛中蛋白的最佳超声提取工艺条件。方法:以蜘蛛粉为原料,探究超声处理方法提取蜘蛛水溶蛋白的提取条件,以水提液中蛋白的含量为考察指标,分别对超声处理过程中的提取时间、料液比、PH进行单因素实验,并通过正交试验法优化蛋白超声提取工艺。结果:液料比、提取时间、提取液p H均对蜘蛛的蛋白质提取具有极显著的作用。蜘蛛蛋白的最佳超声提取工艺为料液比1:40 (g:ml),提取时间50分钟,提取液p H10,最佳工艺下,蜘蛛水溶性总蛋白提取率11. 02%。结论:超声波提取蜘蛛水溶性总蛋白具有操作方便,经济高效的特点,在中药成分提取具有很好的应用前景。
Objective: To explore the optimal ultrasonic extraction conditions of water-soluble proteins from spider. Methods:Using spider powder as raw material,to explore extraction conditions of spider water-soluble proteins by ultrasonic treatment. The content of protein in water extract were regarded as an index. The single factor experiments were done to study the effect of the extraction time,the ratio of material and liquid and PH respectively in the process of ultrasonic treatment. The extraction process of protein ultrasonic was optimized by orthogonal test. Results: The test results showed that all three factors( the ratio of liquid and material,the time of extraction and the p H of the extract) had significant effects on the protein extraction of spiders. and its optimal ultrasonic extraction process as follow: the solid-liquid ratio of 1: 20,the extracting time of 50 min,and p H value of 10. Extraction rate of spiders water-soluble total protein were 11. 02% under the optimum condition. Conclusion: Ultrasonic extraction of water soluble total protein of spider has the characteristics of convenient operation and economic efficiency,which has extensive application prospect in extraction of traditional Chinese medicine.
