摘要
目的优化和建立川牛膝多糖的提取工艺。方法以苯酚-硫酸法测定多糖含量,通过L9(34)正交试验,以料液比、复合酶(纤维素酶:果胶酶:蛋白酶=1:1:1)用量、酶解p H、酶解温度为影响因素,多糖提取率为指标优化提取工艺。结果最佳提取工艺为料液比为1:20(g/ml)、复合酶用量为0.7%、p H值为6.0、温度为45℃。结论该提取工艺周期短、提取率高、简便,易行,为后期多糖研究奠定基础。
Objective The content of polysaccharide was determined by phenol - sulfuric acid method. And the extraction process of Polysaccharides from Achyranthes bidentata was optimized by L9 (34) orthogonal test. Methods Through the L9 (34) orthogonal test, the extraction process of polysaccharide was optimized by the ratio of material to liquid, compound enzyme (cellulase: pectinase: protease =1:1:1), enzymatic hydrolysis of pH, and temperature of enzymolysis. Results The optimum extraction process is:the ratio of material to liquid is 1:20 (g/ml), the dosage of composite enzyme is 0.7%, pH value is 6, and the temperature is 45 degrees. Conclusion The extraction process is short,the extraction rate is high, simple and feasible,and lay the foundation for later polysaccharide research.
