乌金猪、青峪猪和成华猪免疫器官多种免疫调节基因表达的比较 被引量:2

Comparison of multiple immunoregulatory gene expressions in the immune organs of Wujin,Qingyu and Chenghua pigs

摘要 为了阐明乌金猪、青峪猪和成华猪这些地方猪种繁殖力高、抗病耐逆性强等分子免疫特征,采用实时荧光定量PCR方法,研究乌金猪、青峪猪、成华猪与约克夏猪的胸腺、脾脏、扁桃体、肠系膜淋巴结和肺门淋巴结中的选择素-L(CD62L)、转化生长因子-β1(TGF-β1)和白细胞介素(IL-2、IL-4、IL-6、IL-7、IL-12、IL-15)mRNA的表达差异。结果显示:成华猪胸腺IL-2、IL-12、IL-6、IL-7、CD62L,脾脏IL-15,扁桃体CD62L,肠系膜淋巴结IL-2,肺门淋巴结TGF-β1 mRNA表达量均显著高于其他猪种(P〈0.05);乌金猪胸腺TGF-β1,脾脏CD62L,扁桃体IL-6、IL-7,肠系膜淋巴结IL-4 mRNA表达量显著高于其他猪种(P〈0.05);青峪猪肺门淋巴结IL-4、CD62L mRNA表达量显著高于其他猪种(P〈0.05)。研究结果为培育抗病力强的动物新品种和筛选抗病分子标记提供科学依据。 In order to clarify the molecular immunal characteristics of Wujin pigs,Qinglan pigs and Chenghua pigs with high fecundity,strong disease resistance and stress tolerance,fluorescent quantitative PCR was used to analyze the expressions of Selectin-L( CD62L),transforming growth factor-β1( TGF-β1),and interleukin( IL-2,IL-4,IL-6) IL-7,IL-12,IL-15) mRNA in thymus,spleen,tonsil,mesenteric lymph nodes,and hilar lymph nodes of those pigs. The results showed that,compared to the control pigs,the expression levels IL-2,IL-12,IL-6,IL-7,CD62 L in thymus,IL-15 in spleen,CD62 L in tonsils,IL-2 in mesenteric lymph nodes,and TGF-β1 mRNA in hilar lymph nodes of Chenghua pigs were significantly higher than that in other pig breeds( P〈0. 05),that the expression levels TGF-β1in the thymus,CD62 L in the spleen,IL-6 and IL-7 in the tonsil,IL-4 in the mesenteric lymph nodes of Wujin pigs were significantly higher than that in other pig breeds( P〈0. 05),and that the expression levels of IL-4 and CD62 L mRNA in the hilar lymph nodes of Qingyu pigs were significantly higher than that in other pig breeds( P〈0. 05). The present study provides a scientific basis for hybrid breeding of new disease-resistant animal species and screening for breeding molecular markers.
