摘要
目的:考察在不同灌装温度和存储时间下,中药汤剂包装材料中邻苯二甲酸二-(2-乙基)己酯(DEHP)迁移量的变化。方法:采用HPLC法测定DEHP的含量,色谱柱为Cosmosil-C18色谱柱(150mm×4.6mm,5μm),流动相为甲醇-乙腈-水(10∶90∶0.5),流速1.0ml/min,柱温30℃,检测波长224nm。测定DEHP的重复性、精密度和加样回收率,在40、60、80、100℃灌装的中药汤剂,保存1、3、7、14d后测定其中DEHP的含量。结果:DEHP在0.266~8.5mg/L浓度范围内与峰面积呈良好线性关系(r=0.999 5),加样回收率为(99.86±0.84)%(n=6)。在不同灌装温度下,保存1、3、7、14d后检测DEHP的迁移量:40℃灌装时分别为(18.00±0.09)、(21.00±0.06)、(50.00±0.24)、(127.00±0.13)μg/kg;60℃灌装时迁移量分别为(36.00±0.20)、(34.00±0.19)、(57.00±0.13)、(102.00±0.37)μg/kg;80℃灌装时迁移量分别为(43.00±0.25)、(44.00±0.12)、(45.00±0.14)、(43.00±0.23)μg/kg;100℃灌装时迁移量分别为(45.00±0.11)、(44.00±0.14)、(31.00±0.20)、(31.00±0.07)μg/kg。40~100℃灌装的中药汤剂保存1~14d,按每包汤剂200ml,每天服用2包计算,DEHP摄入量为7.2~50.8μg/d。结论:中药汤剂的最优灌装温度为80℃。
Objective: To investigate the migration amount of plasticizer di (2 ethylhexyl) phthalate (DEHP) in the packaging material of Chinese herbal decoction at different packaging temperatures and storage time .Methods : The concentra tion of DEHP was determined by HPLC method ,with Cosmosil C18 column (150 min×4.6 mm ,5 μm),methanol acetonitrile water (10 : 90 : 0.5 ) as the mobile phase at a flow rate of 1.0 ml/min .The column temperature was 30℃ and the detection wavelength was 224 nm .The repeatability ,precision and reeovmT rates of DEHP were determined .The Chinese herbal decoc tion was packaged at 40,60,80 and 100℃, respectively ,and the contents of DEHP were determined after storage for 1,3,7 and 14 days .Results: DEHP was in good linearity with its peak area within the range of 0.266 8.5 mg/L(r=0.999 5).The recovery rate was (99.86±0.84)% (n 6) .When the decoction was stored for 1,3,7,14 days ,the migration amounts of DEPH at different temperatures were : (18.00±0.09) ,(21.00±0.06 ) ,(50.00±0.24) and (127.00±0.13)μg/kg at 40℃; (36.00± 0.20) ,(34.00±0.19) ,(57.00±0.13) and (102.00±0.37) μg/kg at 60 ℃ ;(43.00±0.25) ,(44.00±0.12 ) ,(45.00±0.14) and (43.00 ± 0.23) tzg/kg at 80℃; (45.00 ± 0.11), (44. O0 ± O. 14), (31.00± 0. 20) and (31.00± 0.07) μg/kg at 100 ℃ ,respectively .When the Chinese herbal decoction was packaged at 40 100 ℃ and stored for 1-14 days, the intake amount of DEHP was 7.2 50.8 μg/d calculated by 200 ml per pack and 2 packs per day .Conclusion : The optimal packaging temperature of Chinese herbal decoction is 80 ℃.
