摘要
目的探讨脓毒症大鼠早期应用美托洛尔后对心肌的保护作用。方法将54只健康雄性SD大鼠随机分成3组:假手术组、脓毒症组、美托洛尔组。各组造模3、6、18 h后麻醉处死大鼠,用酶联免疫吸附法测血清心肌肌钙蛋白(cTnI)、炎症介质肿瘤坏死因子α(TNF-α)、白介素1β(IL-1β),留取完整大鼠的左室心肌组织行病理切片,观察心脏损伤情况。结果美托洛尔组各时间点血清cTnI、TNF-α、IL-1β水平均高于假手术组,但明显低于脓毒症组(P <0. 05);脓毒症组大鼠心肌组织病理形态和超微结构损伤明显,美托洛尔可明显保护脓毒症大鼠心肌病理形态和超微结构完整性。结论美托洛尔对早期脓毒症大鼠的心肌具有保护作用,可能与减少炎症介质释放有关。
Objective To investigate the effects and mechanisms of metoprolol on the myocardial damage in rats with sepsis early. Methods 54 rats were randomly divided into slam group （ n= 18 ） , sepsis group （ n = 18 ） and metoprolol group （ n = 18 ）. The rats were killed after 3 h,6h and 18h after anesthesia, and the level of serum cTnI, TNF - ct, IL - 1β were measured by enzyme linked im- munosorbent assay （ELISA）. The left ventrieular myocardium of the intact rats was taken for pathological section and the cardiac damage was observed. Results Serum levels of cTnI, TNF - ct, IL - 1βin metoprolol group were higher than those in sham group but lower than those in sepsis group （ P 〈 0.05 ）. The pathological morphology and ultrastructural injury of myocardium in the sepsis group were obvi- ous, and metoprolol could obviously protect the pathological morphology and ultrastructural integrity of myocardium in the sepsis group. Conclusion Metoprolol has protective effect on myocardium of early sepsis rats, which may be related to decreasing catecholamine sen- sitivity and inflammatory mediators released.
