摘要
茉莉酸是植物响应生物和非生物逆境胁迫的关键激素之一,而茉莉酸信号受体蛋白(Coronatine-insensitive protein 1, COI1)在茉莉酸信号转导过程中发挥着关键作用。本研究基于海南龙血树转录数据中的转录本序列,利用注释拼接和RT-PCR的方法首次鉴定了1个编码海南龙血树茉莉酸受体COI1基因(命名为DcCOI1, GenBank登录号为MF193604),DcCOI1基因长度为2 274 bp,包含一个1 785 bp的完整开放阅读框,编码594个氨基酸,Dc COI1蛋白与芦笋COI1序列的一致性为94%,系统进化分析中与芦笋、剑兰划为同一分支,同属于单子叶植物大类。生物信息学分析显示,其编码蛋白包含COI1的蛋白的F-box特征结构域,分子量为64.14 k D,理论等电点为6.09,为非分泌型蛋白,且不含跨膜结构域,包含52个磷酸化位点,主要在细胞质中发挥生理作用;二级结构主要有α螺旋(43.10%)和无规则卷曲(29.63%)构成。实时荧光定量PCR检测发现DcCOI1在海南龙血树根、茎、叶和花中均有表达,根和茎中的表达量较高,而花中的表达量最低。此结果为阐明海南龙血树血竭形成过程中茉莉酸的调控作用研究奠定了基础。
Jasmonic acid (JA) is one of most important hormone in plant interactive with (a) biotic stress. Coron- atine-insensitive protein 1 (COI), as the receptor of JA signal, play the key roles in JA signaling transduction. Base on the transcriptome database from our previous study, one COIl gene contained completed code sequence in Dracaena cambodiana is identified with annotation and assembly of sequence method and RT-PCR technology, named DcCOI1. Its accession number in GenBank is MF193604. The length of DcCOI1 is 2274 base pairs and its complete open reading frame is 1 785 base pairs which encoding 574 amino acid. The protein homology comparison revealed that DcCOI1 protein is in 94% similarity with the COIl of Asparagus officinali, and DcCOIl is classified in the same branch within Asparagus officinali and Gladiolus hybrid which were all divided into Liliales of monocots. The bioinformatics analysis showed that the molecular weight and isoelectric points of DcCOI1 were respectively 64.14 kD and 6.09, and contained a special F-box motif. DcCOI1 was a non-secretory protein and included 52 phosphorylation sites without transmembrane domain. This protein played the majorphysiological roles in cytoplasm. The secondary structure predicted that DcCOI1 α-helix accounted for 43.10% and random coil for 29.63%. The tissues special expression profiles of De COH in D. eambodiana with real-time fluorescence quantitative PCR indicated that its relative expression in root was higher than other tissues. This study laid the foundation for revealing regulating mechanism ofjasmonic acid in dragon's blood formation from D. cambodiana.
