摘要
目的优选牡丹皮中丹皮酚的提取及包合工艺。方法以丹皮酚含量为考察指标,对水蒸气蒸馏法提取丹皮酚的加水倍量、馏出液量进行动态考察;以丹皮酚包合率为考察指标,以β-环糊精(β-cyclodextrin,β-CD)与丹皮酚的比例、β-CD与加水量的比例、包合时间为考察因素,利用正交设计优化球磨法制备丹皮酚包合物的工艺。结果牡丹皮中丹皮酚的最佳提取工艺为12倍水收集8倍量馏分,冷藏,析晶,抽滤,干燥;球磨法制备丹皮酚包合物的工艺为:取丹皮酚结晶置于球磨机罐中,适量95%乙醇溶解,β-CD与丹皮酚的比例为8∶1(W/W)、β-CD与加水量的比例为1∶3(W/V)、包合时间为60分钟。结论优选的方法简便可靠,适用于牡丹皮中丹皮酚的提取及包合物的制备。
Objective To optimize the extraction and inclusion technology of paeonol from Cortex Moutan Radicis . Methods The transfer rate of paeonol was selected as the evaluation index to study the amount of water and the amount of distillate in extraction of paeonol with the method of steam distillation. With the paeonol encapsulation efficiency as the evaluation index, the inclusion of paeonol with β-cyclodextrin (β-CD) was studied by the ratio of β-CD and paeonol, the ratio of β-CD and water and inclusion time as study factors and by ball grinding method and orthogonal test. Results The steps of optimum extraction technology of Cortex Moutan Radicis was adding 12 times of water and collecting 8 times of distillate, cold storage, crystallization, suction filtration, desiccation, paeonol was obtained. The ball grinding method and orthogonal test technology referred to the solution of paeonol crystal in ball mill pot with 95% ethanol. The optimum inclusion of paeonol with β-CD was that the ratio of β-CD and paeonol was at 8∶1 (W/W), the ratio of β-CD and water was at 1∶3 (W/V) and inclusion for 60 minutes. Conclusion The optimized technology is simple and reliable and fit for the paeonal extraction and inclusion of Cortex Moutan Radicis .
