摘要
目的:研究不同采收月份女贞子中红景天苷和特女贞苷的含量变化规律,为确定女贞子的最佳采收时间提供依据。方法:女贞子样品以60%甲醇超声提取30 min,采用高效液相色谱法测定,色谱条件:ACE Excel 5 C18-AR色谱柱(4.6 mm×250 mm, 5.0μm),流动相:乙腈(A)-水(B),梯度洗脱,检测波长:275 nm,柱温:35℃,流速:1.0 mL·min-1。结果:红景天苷和特女贞苷分别在0.023,74~0.593,6μg(R2=0.999,8)、0.115,8~2.895μg(R2=0.999,9)内线性关系良好,不同采收月份的女贞子随着特女贞苷含量逐渐升高(12月含量最高),红景天苷含量不断下降(9月含量最高)。结论:女贞子的生长过程中可能存在"一分子红景天苷和一分子裂环环烯醚萜转化为一分子特女贞苷"的生物代谢途径,综合考虑红景天苷和特女贞苷总含量变化趋势,建议12月作为株洲女贞子药材的适宜采收期。
Objective: To study on content change tendency of salidroside and specnuezhenide in Nvzhenzi(Ligustri lucidi fructus) at different harvest time, then to provide the experimental basis for determining the reasonable harvest time. Methods: The sample of Ligustri lucidi fructus was extracted 30 min by ultrasonic with60% methanol; The extractive was determined by HPLC, the analytical column was ACE Excel 5 C18-AR(4.6 mm×150 mm, 5.0 μm); The mobile phase was composed of acetonitrile(A) and water(B) with gradient elution; The detection wavelength was 275 nm; The column temperature was set at 35 ℃ and the flow rate was 1.0 mL/min.Results: The linear ranges were in the range of 0.02374-0.5936 μg(R2=0.9998), 0.1158-2.895 μg(R2=0.9999) for salidroside and specnuezhenide. The content of salidroside was decreasing with the increase of the content of the specnuezhenide in Zhuzhou. Conclusion: The author speculate that there is a biological metabolic pathway "A molecular salidroside and a molecular secoiridoid were converted into a molecular specnuezhenide" exists in the growth of Ligustri lucidi fructus. Comprehensive consideration of the total content change tendency of salidroside and specnuezhenide, the December is suggested to be the reasonable harvest time for Ligusri lucidi fructus in Zhuzhou.
