摘要
目的观察冬凌草提取物对SD大鼠是否具有母体、胚胎和致畸毒性。方法受孕成功的雌鼠按体质量随机分为冬凌草低、中、高剂量组(分别为2.5 g/kg BW、5.0 g/kg BW、10.0 g/kg BW)和阴性对照组(相应容量蒸馏水)。在受孕第6 d^15 d,每天灌胃给予受试物,试验期间记录大鼠孕期体质量,并于孕期第20 d处死母鼠,剖腹取出子宫称量子宫连胎重,检查并记录黄体数、活胎数、吸收胎数及死胎数。同时记录活胎仔的体长、体质量以及性别,检查活胎仔的外观、骨骼和内脏的发育情况。结果各剂量组的各项指标与阴性对照组比较,差异无统计学意义(P>0.05)。结论在本实验条件下,冬凌草对SD大鼠无母体毒性、胚胎毒性和致畸毒性。
Objective To evaluate maternal toxicity,embryotoxicity and teratogenicity of Rabdosia rubescens extract in SD rats.Methods Pregnant rats were randomly divided into three Rabdosia rubescens dose groups( 2. 5 g/kg BW,5. 0 g/kg BW,10. 0 g/kg BW) and one negative control group( distilled water at the same volume). During the 6 thto the 15 thday of gestation,rats in dose groups were daily treated with test substance by gavage respectively. In the course of the study,clinical manifestations and body weight of the pregnant rats were recorded. On the 20 thday in pregnancy,the rats were euthanized and examined for indexes including the weight of uterus with embryo,the number of corpora luteum and the number of live,dead,and reabsorbed fetuses. Then fetuses were removed and their count,weight,sex,length,visceral and skeletal changes were recorded.Results There was no statistical significance on the difference of the indexes between each dose group and the negative control group( P〉 0. 05). Conclusion Under this experimental condition,Rabdosia rubescens did not cause maternal toxicity,embryonic toxicity and teratogenicity in SD rats.
