摘要 目的:探讨冬凌草甲素对LOVO细胞凋亡的诱导及其对P53表达的影响。方法:应用CCK-8法检测LOVO细胞生长抑制率,用流式细胞术检测LOVO细胞的凋亡率和P53的表达。结果:冬凌甲素对LOVO细胞生长抑制呈量效和时效的关系。其作用浓度为1.5、5和8μmol/ml时,LOVO细胞凋亡率分别为17.4%,26.6%和35.8%,与对照组比较有差异(P<0.05,P<0.01);P53的表达分别为48.4%,44.6%和17.4%,与对照组比较,差异有显著性(P<0.01)。结论:冬凌草甲素对LOVO细胞的生长有抑制作用,可诱导结肠癌细胞的凋亡并且和P53的调控有关。 Objective:To investigate the effects of oridonin-induced apoptosis and expression of P53 on LOVO cells.Methods:Cell proliferation of oridonin on LOVO cell was determined by CCK-8 assay.Apoptosis rate and expression of P53 were analysed by Flow Cytometry(FCM).ResuIts:CCK-8 assay showed that oridonin had a moderate anti-proliferation effect on LOVO cells in a time-dependant and dose-dependant manner.LOVO cells treated with 8,5 and 1.5 μmol/ml for 48 hours and the apoptosis rate were 35.8%,26.6% and 17.4% respectively(P<0.05,P<0.01).The expression of P53 were 48.4%,44.6% and17.4% respectively(P<0.01).Conclusion: Oridonin had obvious inhibitory effects on LOVO cells in a dosedependant manner.Oridonin apoptosis-induced related with P53.
