摘要
为寻找植物源天然抗氧化剂,采用单因素试验、正交试验、葡聚糖凝胶柱层析纯化等方法对插田泡叶中SOD酶进行了研究,并对SOD酶的种类进行了初步鉴定。结果显示,插田泡叶中SOD酶的最佳提取条件是:插田泡叶与PBS物料比1︰3(g/mL)、PBS溶液pH 7.6、除杂饱和硫酸铵浓度45%、沉淀SOD饱和硫酸铵浓度90%。此条件下,SOD酶活为720.16 U。SOD粗酶液经葡聚糖凝胶柱层析纯化样品,初步鉴定为Mn-SOD。结果证实,插田泡叶含有SOD酶,这可为SOD新资源的开发及插田泡资源的综合利用提供科学依据。
For finding the natural anti-oxidants, purification and identification of superoxide dismutase from Rubus coreanusMaq. leaves were completed by the methods of one-factor-at-a-time experiments, orthogonal array design and sephadex column chromatography, et al. The results showed that the optimal extraction conditions were material/liquid ratio of 1:3(g/mL),PBS solution(pH 7.6), the concentration 45% of saturated ammonium sulfate for removing impurities and the concentration 90% of saturated ammonium sulfate for precipitation SOD protein. Under these conditions, the enzyme activity of SOD from Rubus coreanus Maq. leaves was 720.16 U. The purification sample by the sephadex column chromatography was indicated as Mn-SOD. In summary, Rubus coreanus Maq. leaves contain Mn-SOD. It could provide a scientific basis for the development of the new SOD resources and the comprehensive utilization of Rubus coreanus Maq. resource.
