褪黑激素对慢性应激性抑郁症大鼠行为及血清白细胞介素-2、白细胞介素-6水平的影响 被引量:2

摘要 目的观察褪黑激素(MT)对慢性应激性抑郁症大鼠行为及血清白细胞介素(IL)-2和IL-6水平的影响。方法 42只SD大鼠随机分为空白对照组、模型组和MT治疗组,每组14只。模型组和MT治疗组采用慢性不可预见性轻度应激建立抑郁症大鼠模型,MT治疗组于应激第1天开始每天腹腔注射MT,其余两组每天腹腔注射生理盐水。比较3组大鼠应激前及应激后28 d体重、糖水偏爱指数和强迫游泳静止时间,以及应激后28 d血清IL-2、IL-6水平。结果应激28 d后,模型组大鼠体重、糖水偏爱指数及血清IL-6水平均低于空白对照组及MT治疗组(均P<0.05),应激第14、21、28天,模型组大鼠体重增加量均低于空白对照组及MT治疗组(均P<0.05);而MT治疗组与空白对照组上述指标比较,差异均无统计学意义(均P>0.05)。模型组、MT治疗组、空白对照组大鼠强迫游泳静止时间依次缩短(P<0.05)。结论 MT可明显改善慢性应激性抑郁症大鼠的抑郁行为,其可能是通过降低血清IL-6水平而发挥抗抑郁作用。 Objective To observe the effects of melatonin(MT) on behavior and serum interleukin(IL) -2 and IL45 levels in the rats with chronic stress depression. Methods Forty-two SD rats were randomly divided into blank control group, model group and MT treatment group, with 14 rats in each group. The model group and MT treatment group used chronic unpredictable nfild stress to establish the rat model of depression. The MT treatment group received intraperitoneal injection of MT every day from the first day of stress, and the other two groups were intraperitoneally injected with normal saline every day. Body weight, sugar water preference index and forced swimming rest time were compared among the three groups before stress and after 28 days of stress ,and the levels of serum IL-2 and IL-6 after 28 days of stress were compared as well. Results After 28 days of stress,the body weight, sugar water preference index and serum IL-6 level of the rats in the model group were lower than those in the blank control group and MT treatment group( all P 〈0.05 ) ,on the 14th ,21st and 28th day of stress,the increment of body weight of the rats in the model group was lower than that in the blank control group and MT treatment group ( all P 〈 0.05 ) ; there was no significant difference between the MT treatment group and the blank control group in the indices above ( all P 〉 0.05 ). The forced swimming rest time in the model group, MT treatment group and blank control group decreased in turn ( P 〈 0. 05 ). Conclusion MT can significantly improve the depressive behavior of rats with chronic stress depression, which may play an antidepressant role by lowering serum IL-6 level.
