摘要
目的:研究电针对围绝经期抑郁症大鼠海马神经元凋亡的影响,探讨其治疗抑郁症的作用机制。方法:将48只雌性SD大鼠随机分为空白组、模型组、药物组和电针组各12只;采用去势和慢性不可预见性应激制备围绝经期抑郁症大鼠模型,药物组采用盐酸氯米帕明灌胃,电针组选择"百会""三阴交"和"肾俞"。检测阴道图片,比较大鼠强迫游泳不动时间,采用ELISA法检测大鼠海马DA、5-HT和Caspase-3含量,通过尼氏染色法观察DG区神经细胞的病理变化。结果:与模型组比较,电针组和药物组强迫游泳不动时间明显缩短;海马DA和5-HT含量升高,Caspase-3蛋白表达降低,DG区神经细胞数目增多,胞浆内尼氏小体总面积、积分光密度和平均黑度均升高,差异有统计学意义。结论:电针疗法明显改善大鼠的抑郁症状,阻止神经细胞的凋亡,促进大鼠海马神经细胞的修复和新生。
Objective: To evaluate the effect of electroacupuncture(EA) therapy on neuronal apoptosis in the hippocampal of depression rat model,and investigate its pharmacological mechanisms in depression treatment.Methods:Forty-eight male SD rats were randomly divided into 4 groups: control group,model group,medicine group and EA group.The rat model was established under bilateral overiectomy combined with chronic unpredictable stress(CUMS).Medicine group was treated with clomipramine hydrochloride orally and EA group received EA therapy at Bai hui(DU20),San yin jiao(SP6) and Shen shu(BL23).The behavior was measured by forced swimming test.We detected vaginal pictures,dopamine(DA),serotonin(5-HT) and cysteinyl aspartate specific proteinase-3(Caspase-3) in the hippocampal by immunoassay(ELISA).Using Nissl's staining method to observe the pathological changes of nerve cells in the dentate gyrus(DG) of hippocampal.Results: Compared with model group,in the forced swimming test immobility time significantly shortened; DA and 5-HT increased,the protein level of Caspase-3 decreased,number of nerve cells,total area,integral optical density(IOD) and average blackness of nissl 's body in DG area increased,the difference was statistically significant.Conclusion: Electroacupuncture therapy can reduce the depression symptoms and reverse neurocyte apoptosis in the depression rat model,help to increase hippocampal neuron generation,survival and neogenesis.
