摘要
目的探讨基因表达汇编(GEO)数据库中膀胱癌组织的极光激酶B(AURKB)基因的表达情况及其与膀胱癌临床病理特征和预后的关系。方法收集NCBI的GEO数据库的膀胱肿瘤公共数据集,下载膀胱癌组织AURKB表达数据和临床病理参数。分析AURKB表达与膀胱癌临床病理特征(性别、年龄、侵袭性、T分期、N分期、M分期和疾病分级)及复发转移和预后的关系,利用基因富集分析(GSEA)法分析受AURKB调控的相关基因。结果 AURKB在膀胱癌组织中的表达水平为9.621±0.085,高于正常膀胱组织的7.691±0.059,差异有统计学意义(P<0.05);膀胱癌组织AURKB表达与性别、侵袭性、T分期、疾病分级和进展有关(P<0.05),与年龄、N分期、M分期和复发无关(P>0.05);AURKB高表达组的5年总生存率和5年肿瘤特异性生存率分别为52.226%和70.256%,均低于低表达组的70.112%和90.687%,差异有统计学意义(P<0.05)。GSEA结果显示AURKB高表达样本富集了有丝分裂、E2F转录因子、G2M检查点、MYC信号通路、未折叠蛋白反应和有丝分裂纺锤体相关的基因集。结论 AURKB在膀胱癌中高表达,与膀胱癌的多个临床病理特征相关,且有作为判断膀胱癌患者预后的标志物和治疗分子靶标的潜能。
Objective To investigate the expression of the aurora kinase B (AURKB) gene in the bladder cancer tissues inthe NCBI Gene Expression Omnibus (GEO) database and its relationship with the clinicopathological features and prognosis of bladdercancer. Methods GEO datasets were collected and expression profile of AURKB and clinicopathological parameters of bladder cancertissues were download. The relationship between the AURKB expression and the clinicopathological parameters of bladder cancer (sex,age, invasiveness, T staging, N staging, M staging and disease classification) as well as its relationship with recurrence, metastasisand prognosis were analyzed. The genes regulated by AURKB were analyzed by gene enrichment analysis (GSEA). Results The expression level of AURKB in bladder cancer tissues was 9.621±0. 085, higher than 7. 691± 0. 059 of normal bladder tissues, and thedifference was statistically significant (P〈0.05). The expression of AURKB in bladder cancer was related to sex, invasiveness, T stag.ing, disease classification and progression (P〈0.05), not related to age, N staging, M staging and recurrence (P〉0.05). The 5.yearoverall survival rate and 5.year tumor specific survival rate in the high.expression group of AURKB were 52. 226% and 70. 256%, low.er than 70. 112% and 90. 687% in the low.expression group, and the difference was statistically significant (P〈0. 05). GSEA resultsshowed that AURKB high.expression samples enriched the mitosis, E2F transcription factors, G2M checkpoints, MYC signaling path.ways, unfolded protein responses and mitotic spindle related genes. Conclusion AURKB was highly expressed in bladder cancer andcould be considered as a potential marker and target in the diagnosis and treatment of bladder cancer.
