Nek7与NLRP3炎性小体激活机制的研究进展 被引量:6

The mechanism of Nek7 involvement in the NLRP3 inflammasome activation

摘要 目前,Nod样受体蛋白3(nod-like receptor protein 3,NLRP3)炎性小体在免疫与人类疾病中的重要性已经得到公认,但NLRP3炎性小体的具体激活机制尚不清楚。NIMA相关蛋白激酶7(NIMA-related kinase 7,Nek7)参与NLRP3炎性小体激活,最终导致白介素IL-1β、IL-18的成熟及分泌。同时Nek7也被证实参与有丝分裂纺锤体形成和中心体的分离,且发现NLRP3炎性小体的激活和有丝分裂不能同时发生。因此,Nek7可能作为有丝分裂和NLRP3炎性小体激活两者之间的转换。本文就Nek7及NLRP3炎性小体的激活关系做一综述,以期为炎症性疾病提供新的治疗方向。 At present, the importance of the NLRP3 inflammasome in the immunization and human disease has been recognized. But the mechanism and regulation of NLRP3 inflammasome activation is unclear. In recent years, studies had reported that Nek7 was involved in the activation of NLRP3 inflammasome, eventually leading to the maturation and secretion of IL-1β and 1 L-18. NIMA-related protein kinase 7(NIMA-related kinase 7, Nek7) is also involved in the mitotic spindle formation and separation of centrosome, activation and mitosis of NLRP3 inflammasome can not occur simultaneously. Therefore, Nek7, as the switch, which controls the progress of the mitotic and activation of the NLRP3 inflammasome is extremely important. This article reviews the relationships between the Nek7 and NLRP3 inflammasome activation, providing a new way to treat the inflammatory diseases.
