摘要
目的探究柚皮素对脂多糖诱导的肾小球系膜细胞株(HMCL)凋亡的影响。方法将HMCL细胞随机分为HMCL组、脂多糖组、脂多糖+柚皮素低浓度组、脂多糖+柚皮素中浓度组和脂多糖+柚皮素高浓度组。除HMCL组外,其余各组均用脂多糖诱导HMCL细胞凋亡,并分别予不同浓度的柚皮素处理细胞。采用CCK8检测细胞增殖情况,Hoeschet染色和流式细胞仪检测细胞凋亡情况,免疫印迹检测凋亡相关蛋白Caspase-3和Caspase-9的表达水平。结果与HMCL组比较,脂多糖组细胞增殖倍数降低,细胞凋亡率及Caspase-3和Caspase-9的表达水平升高,差异有统计学意义(P<0.01);与脂多糖组比较,不同浓度的脂多糖+柚皮素组细胞增殖倍数显著升高,细胞凋亡率及Caspase-3和Caspase-9的表达水平明显降低,差异有统计学意义(P<0.05)。结论柚皮素通过降低凋亡相关蛋白的表达抑制脂多糖诱导的肾小球系膜细胞凋亡。
Objective To investigate the effects of naringenin (NG) on lipopolysaccharide (LPS)-induced apoptosis of human glomerular mesangial cell line (HMCL). Methods HMCL cells were randomly divided into HMCL group, LPS group, LPS+NG (low concentration) group, LPS+NG (medium concentration) group and LPS+ NG (high concentration) group. Cells in other groups were treated with LPS to induce apoptosis and then treated with different concentrations of NG (5 M, 10 M and 20 M) respectively except HMCL group. Cell proliferation and apoptosis were determined by CCK8 assay, and Hoeschet staining and flow cytometry respectively. The expressions of apoptosis-related proteins (Caspase-3 and Caspase-9) were measured by Western blot. Results Compared with HMCL group, cell proliferation in LPS group was decreased, whereas cell apoptosis rate and the expression levels of Caspase-3 and Caspase-9 were up-regulated markedly in LPS group ( P 〈0.01). Compared with LPS group, the cell proliferation was increased significantly, while cell apoptosis rate and the expression levels of Caspase-3 and Caspase-9 were down-regulated in LPS+NG (5, 10, and 20 M) groups. Conclusion NG inhibits LPS-induced apoptosis of HMCL by inhibiting the expressions of apoptosis-related proteins.
