超声提取红花总黄酮工艺优化及抗氧化活性研究 被引量:13

Optimization of Ultrasonic-assisted Extraction and Antioxidant Activity of Total Flavonoids from Carthamus tinctorius L.

摘要 目的优化红花总黄酮的超声提取工艺,并评价其抗氧化活性。方法采用单因素和正交试验优选最佳工艺,以分光光度法测定含量,并以Vc为对照,通过清除DPPH·和·OH以及还原力试验综合评价其抗氧化活性。结果最佳提取工艺为乙醇浓度70%、料液比1:25(g/ml)、超声温度70℃、超声功率120W、超声时间30min,此条件下提取率为1.10%。红花总黄酮对DPPH·(IC_(50)=8.27mg/L)和·OH(IC_(50)=5.15mg/L)的清除能力较好,还原能力也较强。结论该提取方法简便、可行,提取物具有较强的抗氧化活性。 Objective To optimize the ultrasonic-assisted extraction technology of the total flavonoids from Carthamus tinctorius L. and evaluate the antioxidant activity of the extract. Methods The ultrasonic-assisted extraction technology of total flavonoids in Carthamus tinctorius L. was optimized with single factor and orthogonal experiments,and determinated its content by spectrophotometry. Meanwhile,the antioxidative activity of the extract was evaluated on its ability of scavenging DPPH· and ·OH,total reducing power,with Vc as the control. Results The optimal extraction conditions were obtained as follows: ethanol concentration of 70%,solid-liquid ratio of 1: 25,ultrasonic temperature of 70℃,ultrasonic power of 120 W and ultrasonic time of30 min. Under these conditions,the extraction yield reached up 1. 10%. The extract showed strong DPPH· scavenging activity( IC50= 8. 27 mg/L),·OH scavenging activity( IC50= 5. 15 mg/L) and good total reducing power. Conclusion The extraction method is simple and practicable,and the extract exhibited strong antioxidant activity.
