摘要
目的:研究佩兰乙醇提取物对结肠癌RKO细胞的体外抗肿瘤作用。方法:SRB法检测细胞的生长抑制率;倒置显微镜观察细胞形态的变化;免疫印迹法检测结肠癌RKO细胞中Bcl-2蛋白、Cleaved Caspase-3蛋白的表达情况。结果:佩兰乙醇提取物在3μg/m L时对结肠癌RKO细胞已具有抑制生长作用,而且随着浓度的增加其抑制率也逐渐升高,到20μg/m L浓度时表现为完全抑制。形态学观察结果显示佩兰乙醇提取物处理后结肠癌RKO细胞后,细胞明显皱缩变圆。免疫印迹法检测到佩兰乙醇提取物能降低结肠癌RKO细胞中Bcl-2的表达、增加Cleaved caspase-3的表达。结论:佩兰乙醇提取物对结肠癌RKO细胞具有抑制生长、促进凋亡的作用。
Objective To study the anti - tumor effects of ethanol extract from Eupatorium fortunei Turcz on colon cancer RKO cells in vitro. Methods The growth inhibition rate of cells was measured with SRB assay. The changes of cell morphology were ob- served by inverted microscope. The expression of Bcl - 2 protein and Cleaved Caspase - 3 protein in colon cancer RKO cells were de- tected by Western blot. Results The growth of RKO cells were inhibited of ethanol extract from Eupatorium fortunei Turcz at the con- centration of 3 μg/mL. Its inhibitory rate gradually increased with increasing concentration, and it completely inhibited at the concen- tration of 20 μg/mL. The morphological observation showed that the RKO cells were significantly shrunken and round after the treat- ment with ethanol extract of Eupatorium fortunei Turcz. The result of Western blotting showed that the expression of Bcl -2 was de- creased and the Cleaved caspase - 3 was increased after the RKO cells were treated with ethanol extract of Eupatorium fortunei Turcz. Conclusion Theethanol extract of Eupatorium fortunei Turcz can inhibit the growth and promote apoptosis of RKO cells.
