响应面法优化杏鲍菇液体发酵培养基 被引量:1

Optimization of Pleurotus eryngii Liguid Fermentation Medium by Response Surface Methodology

摘要 设计并通过响应面优化了一种适用于杏鲍菇(Pleurotus eryngii)液体发酵的新培养基TSM-1。以单因子试验筛选出指标具有显著影响的TSD-1微生物生长促进剂,液体多肽蛋白肥和Na Cl,并通过Box-Behnken设计以及响应面分析得到了3因子最优添加量,形成TSM-1培养基的最优配方:葡萄糖40 g,酵母浸膏10 g,KH_2PO_41 g,MgSO_4·7H_4O 0.5 g,NaCl 0.15 g,复合维生素B片1片(市售OTC成药),添加TSD-1微生物生长促进剂0.3 g,钙蛋白2 g,蒸馏水1 000 mL,pH值6.4,121°C灭菌30 min。优化后TSM-1培养基中杏鲍菇菌丝生物量可达18.70±0.03 g·L^(-1)。 A new Pleurotus eryngii liquid culture medium named TSM-1 was designed and optimized by response surface methodology. Three factors were selected by single factors experiment including TSD-1 microbial growth promoter, liquid protein fertilizer and Na Cl. The optimum additive amounts of three factors were determined respectively by Box-Benhnken design and response surface analysis. The optimum formula of TSM-1 culture medium was as follows: glucose 40 g, yeast extractor 10 g, KH_2PO_41 g, MgSO_4·7H_4O0.5 g, NaCl 0.15 g, multivitamin tablet1 piece, adding TSD-1 0.3 g, liquid protein fertilizer 2 g, distilled water 1 000 m L, pH=6.4, steril-ization at 121 ℃ after 30 min. Under the optimized TSM-1 medium, the biomass of Pleurotus eryngii was reached to 18.70±0.03 g·L^(-1).
