不同光敏剂介导光动力疗法对杀菌有效性及牙髓安全性的研究 被引量:4

摘要 目的:探讨甲苯胺蓝(TBO)和亚甲蓝(MB)介导的光动力疗法(PDT)对杀灭大鼠口腔变异链球菌(S.mutans)的有效性及其对牙髓组织安全性的影响。方法:构建大鼠龋损模型44只,根据半导体激光能量密度及光敏剂的不同,将大鼠随机分为6个实验组:4.8、9.6、19.2 J/cm2的TBO-PDT组和MB-PDT组;5个对照组:单纯TBO组,单纯MB组,单纯激光照射组(19.2 J/cm^2),2 g/L Na F组,生理盐水组,共11组(n=4)。选取各大鼠口腔每区最后2个磨牙(共8个牙)牙合面按分组进行相应处理后,用平板菌落计数法计算各组的杀菌率,组织病理学观察牙髓组织的变化。结果:平板菌落计数结果显示,与生理盐水组相比4.8、9.6、19.2 J/cm^2PDT各组对S.mutans均有明显的杀菌效果(P<0.05),杀菌率与能量密度的增长成正比;单纯光敏剂组对S.mutansr杀菌作用则无统计学差异(P>0.05)。组织病理结果显示,激光能量密度升高时,可对大鼠牙髓组织产生不同程度的炎症或出血损伤,其中19.2 J/cm^2单纯激光组的损伤程度最大。结论:PDT对S.mutans的杀菌效果明显,且与光敏剂本身无关,但较高能量密度将对牙髓组织造成损伤,需选择合适的照射能量密度。 AIM: To investigate the effects of methylene blue(MB) and Toluidine blue O(TBO) mediated photodynamic therapy (PDT) on the inhibition of S. mutans and the safety for dental pulp tissue in rats. METHODS : Cariogenic model was established on the bilateral 2 Back molars in 44 Wistar rats and the rats were divided into 11 groups randomly(u =4 rats and 8 teeth). The teeth surfaces were treated by semiconductor laser as a light source with TBO and MB respectively as the photosensitizer. The colony formation of S. mutaus from tooth decay was examined by plate count. The changes of dental pulp tissue and oral soft tissue were observed by histological and pathological exami- nation. RESULTS : Plate count showed that PDT could significantly reduce the olony formation of S. mutaus. With the increase of energy density, the bactericidal rate of caries was increased, but the effects of caries prevention was unrelat- ed with photosensitizer. Histopathological results showed that when the energy density was increased, the different lev- els of dental pulp tissue and oral soft tissue were damaged, the damage degree of TBO - PDT 19.2 J/cm2 group was the most. CONCLUSION: PDT has obvious anti - caries effect, but over a fluence range, PDT can make some injuries to dental pulp tissue and oral soft tissue.
