摘要
苯氧羧酸类除草剂2,4-滴丁酸具有药效高、价格低的优点,在全球被广泛应用于防除禾本科作物的双子叶杂草、莎草、及某些恶性杂草。为从分子层面上考察2,4-滴丁酸与牛血清蛋白之间的相互作用,探究其对人体造成的潜在危害,本试验选择牛血清蛋白作为研究对象,采用紫外-可见光谱(UV-Vis)、荧光光谱(FS)、圆二色光谱(CD)及傅里叶红外光谱(FT-IR)等分析方法。结果表明:一分子的2,4-滴丁酸能够与一分子的牛血清蛋白结合形成复合物,蛋白的内源荧光被猝灭,这个过程主要靠疏水作用力驱动,同时也伴随着蛋白构象的改变,以此判断两者的结合机制属为静态猝灭。研究表明:2,4-滴丁酸具有潜在的环境毒性,同时也为今后评价其在人体的致毒机理提供了理论依据。
As an effective and cheap phenoxy acid herbicide,2,4-D-butyric acid(2,4-DB)is widely used in the control of dicotyledonous weeds,sedges,and certain malignant weeds in the world.To study the potential toxicity mechanism of 2,4-DB,we used bovine serum albumin(BSA)as a model at the molecular level via UV-Vis,fluorescence,circular dichroism and fourier transform infrared spectroscopy.The results indicated that there was only one molecule of 2,4-DB binding to one molecule of BSA.Thus the complex between 2,4-DB and BSA formed and the intrinsic fluorescence of BSA was quenched.Hydrophobic forces played a major role in the binding process of 2,4-DB to BSA.This work can help people see the microscopic poisoning mechanism when 2,4-DB went into environment.
