摘要
目的探讨血必净对脓毒症大鼠IL-6、TNF-α、甲状腺激素水平及下丘脑和脂肪组织2型脱碘酶(D2)相对表达的影响。方法100只成年雄性SD大鼠按随机分为正常组、假手术组、模型组和血必净组,采用盲肠结扎穿孔术(CLP)制备大鼠脓毒症模型。血必净组以5mL/kg于CLP术后1h静脉注射,其余各组大鼠注射等体积生理盐水,12h后再次给予等量血必净静脉注射。观察术后大鼠精神、活动状态及腹腔内情况以评估模型。各组分别于给药后12h、24h腹主动脉取血,检测大鼠血清T_3、T_4、TSH、IL-6及TNF-α水平;同时,提取下丘脑及脂肪组织总RNA,检测其D2mRNA水平。结果血必净注射液可降低脓毒症大鼠血清IL-6、TNF-α水平,并可调节甲状腺激素T_3、T_4、TSH的水平及下丘脑、脂肪组织D2表达水平。结论血必净注射液可通过降低炎性因子水平及调节血清甲状腺激素水平启动脓毒症大鼠体内保护性机制。
Objective To observe the changes and the intervening effects on IL-6, TNF-α, TH levels and the D2 gene expression in septic rats. Methods 100 adult male SD rats were divided into 4 groups： normal group, fake-operation group, model group and Xuebijing group based on the Random Number Table. Septic rats model with the method of cecal ligation and puncture （CLP） was established. Rats in Xuebijing group were given Xuebijing 5 mL/kg by intravenous injection within 1 h and when operation ended in 12 h, when other rats were given equivalent saline. Mental and active status , the abdominal cavity were obsmwed after operation. Blood of each rat was drawn at the time of 12 h, 24 h after giving Xuebijing. Serum T3, T4 and TSH levels were measured with RIA method, and serum IL-6, TNF-α levels were measured with ELISA method, D2 gene expressions in hypothalamus and adipose tissue were checked with the method of qPCR. Results The TH level and D2 gene expression change at the same time of serum IL-6, TNF-α levels changing in CLP septic rats, and this can be intervened by Xuebijing injection. The mechanism may relate with the influences on D2 by the way of effecting inflammatory factors , and then make the effects of regulating serum TH level in CLP septic rats. Conclusion Xuebijing injection can initiate the protective mechanism in CLP septic rats by reducing the level of inflammatory factors and regulating serum thyroid hormone.
