摘要 目的从细胞外基质成分变化研究理冲汤对子宫肌瘤模型大鼠的影响。方法采用雌孕激素双重造模法,制备大鼠子宫肌瘤模型。以放免法、SP免疫组化法和蛋白印迹技术,观察理冲汤对子宫肌瘤模型大鼠细胞外基质成分的影响。结果与正常对照组比较,模型对照组子宫肌瘤大鼠血清层粘连蛋白(laminin,LN)含量明显升高,子宫肌瘤组织细胞外基质主要成分Ⅳ型胶原表达明显增多(P<0.05)。与模型对照组比较,理冲汤高低剂量组血清LN含量、子宫组织Ⅳ型胶原表达显著减少(P<0.05)。结论子宫肌瘤的发生与细胞外基质代谢异常有关,理冲汤通过调控血清LN含量和子宫组织Ⅳ型胶原的表达,进而抑制肌瘤的生长。 Objective To study the effects of Lichong decoction on uterine leiomyoma in rats from the composition changes of extracellular matrix. Methods The uterine leiomyoma model in rats was established by the method of making model of estrogen and progesterone. The radioimmunoassay immunohis- tochemical method and Western blotting were used to observe the effects of Lichong decoction on the composition of extracellular matrix of uterine leiomyoma in rats. Results Compared with the normal control group, the serum laminin (LN) of the model control group of uterine leiomyoma in rats was significantly in- creased( P〈 0. 05 ). The major components IV collagen expression of extracellular matrix of uterine leiomyoma in rats was raised markedly. Compared with the model control group of uterine leiomyoma in rats, high and low dose Lichong decoction group, the serum LN and IV collagen of the uterus was reduced significantly(P〈0. 05 ). Conclusion Uterine leiomyoma is related to metabolic disorders of extracellular matrix. Lichong decoction inhibits the growth of uterine leiomyoma by regulating the serum LN content and IV collagen expression of the uterus.
