鼻咽癌增殖与转移机制荧光检测的研究进展 被引量:3

Research progress of fluorescence detection to mechanisms of proliferation and metastasis of nasopharyngeal carcinoma

摘要 鼻咽癌是一种高转移性的头颈部肿瘤,EB病毒编码潜伏膜蛋白-1(LMP1)被认为与鼻咽癌的发展、侵袭、转移关系密切,但是LMP1诱导鼻咽癌增殖、转移的机制尚不明确。本文对近年鼻咽癌增殖、转移机制方面的研究进展作简要介绍;荧光检测技术作为一种有效技术手段被广泛应用于生命科学,对应用于鼻咽癌增殖、转移机制研究中的荧光检测技术(免疫荧光、双分子荧光互补、荧光共振能量转移)进行详细总结,并分析了各自的优缺点。针对鼻咽癌增殖、转移与肿瘤细胞质膜关系密切的特点,展望了利用全内反射荧光显微镜(TIRF)结合荧光共振能量转移(FRET)技术获取鼻咽癌细胞质膜上LMP1蛋白高分辨率成像,破解鼻咽癌增殖、转移机制的前景。 Nasopharyngeal carcinoma (NPC) is one of the head and neck cancers with strong metastasis feature,and EBV-encoded latent membrane protein 1 (LMP1) is considered to closely connect with de- velopment,invasion and metastasis of NPC. But the mechanisms of proliferation and metastasis of NPC induced by LMP1 are not clear at present. Research progress on the mechanisms of proliferation and me- tastasis of NPC are briefly presented in this article. As an effective technology,various fluorescence de- tection technologies have been widely applied in life sciences. Characteristics of those technologies (e. g, ImmunoFluorescence,Bimolecular Fluorescence Complementation and Fluorescence Resonance Energy Transfer) which are applied in those researches on the mechanisms of proliferation and metastasis of NPC are detailed summarized. Furthermore advantages and disadvantages of those technologies are ana- lyzed,respectively. According to closely connection between the cytoplasmic membrane and proliferation and metastasis of NPC, the Possibilities that utilizing the technology combining with total internal reflec- tion fluorescence and Fluorescence Resonance Energy Transfer to solve the problem about the mecha- nisms of proliferation and metastasis of NPC through high resolution imaging of LMP1 on cytoplasmic membrane of NPC cells are analyzed.
