摘要
通过对鹅卵泡中的颗粒细胞层进行形态学观察和分析,为在颗粒细胞水平深入研究鹅卵泡的选择与发育过程奠定形态学基础。采集处于产蛋期的皖西白鹅大白、F5、F3和F1卵泡,福尔马林固定后制作石蜡切片,H&E法染色,光镜观察颗粒细胞层形态;机械法分离F5卵泡颗粒细胞,提取总RNA,Real-time quantitative PCR(q RT-PCR)检测分析鹅等级卵泡促卵泡素受体(follicule stimulating hormone receptor,FSHR)mRNA、促黄体素受体(luteinizing horomone receptor,LHR)mRNA表达情况。结果表明:鹅卵泡颗粒细胞层厚度随等级卵泡发育表现为先增加后下降;与卵泡等级发育和排卵相关的FSHR mRNA表达伴随卵泡发育先增加后降低,LHR mRNA表达伴随卵泡发育持续增加。
The morphological observation and analysis of the granulosa cell layer in the follicles of geese laid the morphological foundation for the further study of the selection and development of goose follicles at the level of granulosa cells. The large white follicle(LWF), F5, F3 and F1 of Wanxi White Goose were collected in the laying period, which were fixed in the formaldehyde solution. Then paraffin sections were made and stained with HE. The morphology of granular cell layers was observed by light microscopy. The granulosa cells of F5 were isolated and the total RNA was extracted and then FSHR m RNA and LHR m RNA were detected by real-time quantitative PCR(q RT-PCR). The results showed that the thickness of granulosa cell layer in goose follicles first increased and then decreased with hierarchy follicle development. The expression of FSHR m RNA associated with follicular hierarchy development and ovulation increased first and then decreased with follicular hierarchy development. The expression of LHR m RNA was accompanied by a sustained increase in follicular development.
