摘要
目的探讨黄芪总苷对微波辐射致大鼠心脏损伤保护作用。方法 100只雄性SPF级Wistar大鼠随机分为5组:正常对照组(C)、辐射对照组(R)、黄芪总苷低剂量组(L)、黄芪总苷中剂量组(M)和黄芪总苷高剂量组(H)。采用平均功率密度为30 mW/cm^2的微波辐射15 min,辐射当天开始灌胃给予黄芪总苷40 mg/kg/d、80 mg/kg/d、160 mg/kg/d,每日1次,连续给药14 d。于给药后7 d(辐射后7 d)、停药后6 h(辐射后14 d),通过对大鼠心电图、血清中心肌酶及Ca^(2+)、心脏组织学结构及超微结构观察,研究黄芪总苷对微波辐射致心脏损伤的保护作用和剂量的关系。结果 (1)于辐射后7 d,心率和P波波幅下降(P<0.05或P<0.01),血清中LDH和AST活性均显著升高(P<0.05或P<0.01)。辐射后14 d,P、T波波幅明显降低(P<0.05或P<0.01);Ca^(2+)含量显著升高(P<0.01),AST活性仍高于正常对照组(P<0.05)。黄芪总苷中剂量给药组对上述各指标改变的改善效果显著(P<0.05或P<0.01);(2)辐射后7 d,大鼠心肌细胞水肿,肌纤维排列不整,横纹不清,少量细胞核固缩、深染;心肌细胞线粒体肿胀、形态异常,闰盘模糊。黄芪总苷中、高剂量给药组对上述指标改变的改善效果显著,表现为大鼠心肌细胞圆柱状,未见水肿,肌纤维排列整齐,横纹清晰;细胞核位于细胞中央,呈椭圆形,线粒体形态正常,椭圆形,肌原纤维排列整齐,血管周间隙正常,闰盘结构清晰。结论黄芪总苷对30 mW/cm^2微波辐射所致的大鼠心脏功能紊乱和组织结构损伤具有良好的保护作用,最佳有效剂量为80 mg/kg/d。
Objective To investigate the protective effects of Astragalosides on heart injury of rats induced by microwave radiation. Methods 100 male Wistar rats were given microwave radiation with the average power density of 30 m W/cm^2 for 15 min,and treated by filling stomach with 40 mg/kg/d,80 mg/kg/d,160 mg/kg/d Astragalosides once a day for 14 d consecutively. At the time points of 7 d after the beginning of administration( 7 d after radiation),6 h after the end of administration( 14 d after radiation),electrocardiogram,myocardial enzyme and Ca^2+in serum,and cardiac histological structure and ultrastructure were observed to determine the protective effect and optimal dose of Astragalosides on the microwave radiation-induced heart damage. Results 7 d after radiation,heart rate and P-wave amplitude were significantly increased( P0.05 or P〈0.01). 14 d after radiation,T wave amplitude was significantly increased( P〈0.05),P-wave amplitude was still higher than the normal control group. 7 d after radiation,activities of serum LDH,AST were significantly increased( P〈0.05 or P〈0.01). 14 d after radiation,Ca^2+ content was significantly increased( P〈0.01),and AST activity was still higher than the normal control group( P〈0.05). In the Astragalosides groups of middle and high dose,the improvement was significant( P〈0.05 or P〈0.01). 7 d after irradiation,the myocardium of rats was edematous,the muscle fibers were in wavy arrangement,the stripes were blurred,some nuclei were condensed and stained,the mitochondria were swollen or with abnormal morphology,vascular endothelial cell cytoplasmic vesicle increased,perivascular space widened,intercalated disk were blurred. In the Astragalosides groups of middle and high dose,the abnormal changes were significantly improved: myocardial cells were cylindrical and had no edema,the muscle fibers were arranged in order,stripes were,nuclei were located in the center of the cells with oval shapes.
