摘要
目的:通过体内实验揭示BMP2指节肽(BKP)结合后的Ta_2O_5纳米管是否具有促进种植体表面骨整合的能力。方法:以多巴胺为中间介质,将BKP结合于Ta_2O_5纳米管表面,通过接触角测量和X射线光电子能谱检测BKP和Ta_2O_5纳米管的成功结合。将种植体植入新西兰白兔的胫骨内,采用实时定量PCR检测术后1、2、3、4周种植体表面成骨相关基因ALP和Col-I的表达,制作骨-种植体不脱钙磨片,观察术后2、4、8周种植体周围骨的组织学表现。结果:BKP成功结合至Ta2O5纳米管表面。相对于两组对照组,实验组表面基因表达量有显著的提高且在骨-种植体接触面积和新骨形成方面有明显增加(P<0.05)。结论:在Ta_2O_5纳米管表面附着BKP可明显促进种植体骨整合,这种表面处理方法可用于改善钽种植体的临床应用。
Objective: To explore the effects of BMP2 knuckle peptide(BKP), in combination with Ta2O5 nanotubes, on implant osseointegration in vivo. Methods: Polydopamine was immobilized onto Ta2O5 nanotubes as an intermediate layer,and then BKP was conjugated onto the nanotubes. The successful fabrication of BKP was conιrmed by contact angle measurement and X-ray photoelectron spectroscopy. The implants were then inserted into the tibiae of rabbits, the gene expression of alkaline phosphatase and collagen-I was examined by real-time PCR at 1, 2, 3 and 4 weeks after implantation, and the histological features around the implants on non-decalciιed sections were investigated at 2, 4, and 8 weeks after implantation. Results: There was a signiιcant increase in gene expression levels and bone-implant contact of the bone attached to implants with immobilized BKP compared with the other two control groups. Conclusion: The surface functionalization of Ta2O5 nanotubes with BKP was beneιcial for osseointegration and this approach could be further developed to improve tantalum-based implants for various applications.
