摘要
目的:研究桃红四物汤对乳腺浸润性导管癌病例凋亡蛋白Bcl-2、Bax及细胞增殖核抗原Ki-67表达的影响,探讨其抗癌的可能机制。方法:将本院同期收治的60例浸润性乳腺癌患者分成3组,其中20例为对照组,20例行单纯术前化疗的为化疗组,20例给予术前化疗及桃红四物汤的为复合药物组,用免疫组化检测Ki-67蛋白,用流式细胞仪检测Bcl-2、Bax蛋白,并比较Bcl-2/Bax比值。结果:化疗组及复合药物组Ki-67蛋白表达减少,但不存在组间差异;两组均见Bcl-2蛋白下降,Bax蛋白升高,Bcl-2/Bax比值减小,与对照组对比有显著差异,且复合药物组较化疗组效果更为明显。结论:桃红四物汤联合用药可通过下调Bcl-2蛋白,上调Bax蛋白,降低Bcl-2/Bax比值来促进肿瘤细胞凋亡,可能是其抗癌作用的机制之一。
Objective： To study the effects of Taohong Siwu Decoction on protein expressions of Bcl-2,Bax and Ki-67 in invasive breast cancer,and to explore its possible anti-cancer mechanism. Methods： 60 cases of invasive breast cancer were randomly divided into the control group（ n = 20）,the chemotherapy group（ n = 20）,and the combined group（ n = 20）; the chemotherapy group was only given chemotherapy before operation; on which basis,the combined group was also treated with Taohong Siwu Decoction. Protein expression of Ki-67 was detected by immunohistochemistry,and protein expressions of Bcl-2 and Bax were detected by flow cytometry,and the ratio of Bcl-2/Bax was compared in the groups. Results： The protein expression of Ki-67 was decreased in the chemotherapy group and the combined group,but there was no statistical difference in the three groups. The protein expression of Bcl-2 was significantly decreased,and protein expression of Bax was greatly increased,the ratio of Bcl-2/Bax was obviously reduced in the chemotherapy group and the combined group,compared to those in the control group; the changes were more significant in the combined group. Conclusion： Taohong Siwu Decoction combined with chemotherapy can down-regulate Bcl-2 protein,up-regulate Bax protein and decrease the ratio of Bcl-2/Bax to promote apoptosis of the tumor cells,which may be one of the mechanisms of its anti-cancer effect.
