桃红四物汤对乳腺癌血管生成的影响 被引量:13

Effect of Tao Hong Siwu Decoction on Angiogenesis in Patients with Breast Cancer

摘要 【目的】探讨桃红四物汤对乳腺癌血管生成的影响及机理。【方法】将行术前化疗的40例乳腺癌患者随机分为治疗组和对照组。治疗组20例给予桃红四物汤及术前化疗,对照组20例单纯行术前化疗。观察各组患者化疗前及化疗3个疗程后(21 d为1个疗程)肿瘤的微血管密度(MVD)、血管内皮生长因子(VEGF-A)、血管内皮生长因子受体-1(flk-1)的表达变化。【结果】治疗后2组均能降低MVD、VEGF-A的表达,与治疗前比较差异均有统计学意义(P<0.05);治疗后2组比较,治疗组MVD及VEGF-A表达阳性率均显著低于对照组,差异有统计学意义(P<0.05)。治疗后2组亦能降低flk-1表达,但差异无统计学意义(P>0.05),治疗后2组比较差异无统计学意义(P>0.05)。【结论】桃红四物汤能抑制乳腺癌患者肿瘤血管生成,其作用机制可能与抑制VEGF-A及MVD表达有关。 Objective To study the effect of Tao Hong Siwu Decoction on the tumor angiogenesis and to explore its mechanism. Methods Forty breast cancer patients who had received preoperative chemotherapy were equally randomized into two groups. The treatment group was given oral use of Tao Hong Siwu Decoction and preoperative chemotherapy, and the control group was given only preoperative chemotherapy, 21 days constituting one treatment course. The micro-vessle density ( MVD ) , and the expression levels of vascular endothelial growth factor A (VEGF-A) and vascular endothelial growth factor receptor 1 (ilk-1) in the tumor tissue of both groups were observed before chemotherapy and after 3 courses of chemotherapy. Results After treatment, both groups decreased MVD and the expression level of VEGF-A in tumor tissue (P 〈 0. 05 compared with those before treatment), and the difference between the two groups was statistically significant (P 〈 0.05 ). The expression of ilk-1 in the tumor tissue was decreased in both groups, but the difference was insignificant (P 〉 0.05 ). Conclusion Tao Hong Siwu Decoction can inhibit the angiogenesis in the tumor tissue of patients with breast cancer, and its mechanism is associated with the inhibition of VEGF-A expression and with the decrease of MVD.
