摘要
目的观察富亮氨酸α2糖蛋白1(LRG-1)在溃疡性结肠炎(UC)及结直肠癌(CRC)组织中的表达。方法选取40只C57BL/6J雄性小鼠,随机分为对照组,急性、慢性UC组及UC癌变组,每组各10只。除对照组外,采用偶氮氧甲烷(AOM)及葡聚糖硫酸钠法(DSS)建立急、慢性UC及UC癌变模型,采用实时荧光定量PCR(qRTPCR)检测各组LRG-1 mRNA表达。另收集行结肠镜检并取病理活检的正常结肠黏膜组织20例份,UC组织30例份,CRC组织30例份,通过免疫组化法和qRT-PCR法检测各组织中LRG-1 mRNA及蛋白表达。结果与对照组小鼠比较,LRG-1 mRNA表达在急性、慢性UC组、UC癌变组逐渐增高,组间比较有统计学差异(P均<0.05)。人正常结肠黏膜组织、UC及CRC组织中LRG-1 mRNA及蛋白表达逐渐增高,组间比较有统计学差异(P均<0.05)。结论 LRG-1在UC和CRC组织中呈高表达,其可能在UC、CRC发生发展中发挥重要作用。
Objective To investigate the expression and significance of leucine-rich α2 glycoprotein-1（ LRG-1） in the ulcerative colitis（ UC） and colorectal cancer（ CRC） tissues. Methods We randomly divided forty male C57 BL/6 J mice into four groups with 10 in each group： the control group,acute UC group,chronic UC group,and UC-associated cancer group. We induced the acute and chronic UC with dextran sodium sulfate（ DSS）,and carcinogenesis by addition of azoxymethane（ AOM）. We measured the mRNA expression level of LRG-1 by real-time polymerase chain reaction（ qRTPCR） in the colorectal mucosal tissues of mice from four groups. We assessed the mRNA and protein levels of LRG1 by qRT-PCR and immunohistochemistry in the colorectal mucosal tissues obtained from 20 healthy individuals,30 active UC patients,and 30 CRC patients. Results Compared with the control group,the expression level of LRG-1 mRNA in the active UC,chronic UC,and UC-associated cancer group increased progressively. The difference was statistically significant among these four groups（ all P〈0. 05）. Compared with the healthy individuals,the mRNA and protein expression levels of LRG-1 in the active UC and CRC patients increased gradually. The difference was statistically significant among these three groups（ all P〈0. 05）. Conclusion LRG-1 is highly expressed in UC and CRC tissues,and its high expression plays an important role in the occurrence and development of UC and CRC.
