摘要
目的:探讨当归补血汤对S180荷瘤小鼠的抗肿瘤作用机制。方法:将接种S180肉瘤细胞的小鼠分为阴性组、阳性(环磷酰胺,CTX)组、当归补血汤(低、中、高剂量)组。给药10日后,称体质量,取材,记录瘤重,计算抑瘤率、生长抑制率;记录肝脏、脾脏质量并计算指数,采用HE染色方法观察当归补血汤组肿瘤组织细胞的形态学特征,并采用ELISA法检测血清IL-2含量。结果:CTX组、当归补血汤(低、中、高剂量)组抑瘤率分别为84.45%、27.42%、40.00%、38.53%。与阴性对照组相比,给药组抑瘤效果显著(P<0.05,P<0.01);与阴性对照组相比,CTX组肝指数、脾指数明显降低(P<0.05,P<0.01);当归补血汤各组肝指数和脾指数,与CTX组相比明显升高(P<0.05,P<0.01)。HE染色显微镜观察,瘤组织内出现破裂、空洞,部分区域被非瘤组织取而代之,该现象随着剂量的增加而愈加明显。用ELISA法测得,当归补血汤中、高剂量组IL-2显著提高。结论:不同剂量当归补血汤均能抑制S180肉瘤的生长,作用机制很可能与提高血清免疫因子IL-2有关。
Objective： To explore the anti-tumor mechanism of Danggui Buxue Decoction in S180 tumor-bearing mice.Methods： The mice that received the S180 sarcoma cells were randomly divided into the negative control group,the Cyclophosphamide（ CTX） group,and Danggui Buxue groups（ low-dose,medium-dose,and high-dose）. Tumor weight,tumor inhibition rate,growth inhibition rate,and quality indexes of liver and spleen were recorded. Morphology of tumor cells were observed with HE staining after the medication of Danggui Buxue Decoction,serum content of IL-2 was detected with ELISA method. Results： The tumor inhibition rate was 84. 45% in the CTX group,and it was27. 42%,40. 00% and 38. 53% respectively in Danggui Buxue groups of low-dose,medium-dose and high-dose.Compared to the negative control group,the tumor inhibition effect was significant in the medication groups（ P〈0. 05,P〈0. 01）. In terms of the quality indexes of liver and spleen,they were significantly decreased in the CTX group,compared to the negative control group（ P〈0. 05,P〈0. 01）; however,they were significantly increased in Danggui Buxue groups,compared to the CTX group（ P〈0. 05,P〈0. 01）. HE staining results showed that the tumor tissue was bursting and empty,and some part had been replaced by the non-tumor tissue,which showed dose-dependent relation. The level of IL-2 was significantly increased in the Danggui Buxue group of medium-dose and high-dose. Conclusion： Danggui Buxue Decoction can inhibit the growth of S180 tumor-bearing mice with different doses,and its mechanism may be related to the increase of serum immune factor of IL-2.
