摘要
为建立微波辅助蛋白质水解-反相高效液相色谱测定饲料中色氨酸的方法 ,实验将样品经微波碱水解后,应用Syncronis C18色谱柱,以0.05 mol/L KH2PO4溶液-乙腈(90∶10,V/V)为流动相进行洗脱,DAD检测器在220 nm处进行检测,外标法定量。结果表明:色氨酸线性关系良好,相关系数为1.0000,回收率为93.0%~95.5%,相对标准偏差为1.26%~2.72%,检出限为0.025 mg/g。该方法操作简便、快速、灵敏度高、重现性好,适用于批量饲料样品中色氨酸含量的测定。
In order to established the determination of tryptophan in feed by reversed-phase high performance liquid chromatography after microwave-assisted alkaline hydrolysis of protein.The samples after microwave alkaline hydrolysis,separation and quantification was achieved by using Syncronis C18 column,and mobile phase of 0.05 mol/L KH2 PO4 solution-acetonitrile(90∶10,V/V) were used.The detection was carried out with diode array detector(DAD) by absorbance at 220 nm.The external standard calibration curves were used for quantification.There was good linear correlations for tryptophan(R^2=1.0000).Average spike recovery of tryptophan was 93.0% ~ 95.5%,with relative standard deviations 1.26% ~ 2.72%,the limit of detection for tryptophan being 0.025 mg/g.This method was simplicity,rapidity,good reproducibility and high selectivity,and therefore could be used to determine tryptophan in feed.
