摘要
本研究采用小鼠强迫游泳、小鼠悬尾和自主实验对金丝桃苷的抗抑郁作用进行了评价,同时选择5-HT_(1A)受体阻断剂Way100635、5-HT_2受体阻断剂cyproheptadine、5-HT_(2A)受体阻断剂ketanserin进行小鼠悬尾实验探讨金丝桃苷的抗抑郁机制。结果证实金丝桃苷具有确切的抗抑郁活性;5-HT_(2A)、5-HT_2受体阻断剂与金丝桃苷20mg/kg共同给药组与金丝桃苷20mg/kg单独给药组相比,共同给药组能明显延长小鼠悬尾的累计不动时间(P<0.05)。表明金丝桃苷的抗抑郁活性与5-羟色胺能系统(5-HT_(2A)、5-HT_2受体)有关。
In the present study,the antidepressant activity of hyperoside was evaluated by using forced swimming,mouse suspension and autonomous experiments in mice.At the same time,5-HT1Areceptor blockers Way100635 and 5-HT2receptor blockers cyproheptadine and 5-HT2Awere selected to investigate the antidepressant mechanism of hyperoside by using mouse suspension test.The results confirmed that hyperoside has definite antidepressant activity;at the same time,5-HT2A,5-HT2receptor blockers ketanserin(5mg/kg),cyproheptadine(3mg/kg)and hyperoside after 20mg/kg co-continuous administration,compared with hyperoside 20mg/kg alone,the cumulative immobility time of tail suspension was significantly prolonged(P〈0.05).The hyper-depressant activity of hyperoside has been shown to be related to the serotoninergic system(5-HT2A,5-HT2receptor).
