摘要
目的基于p38丝裂原活化蛋白激酶（p38MAPK）信号通路研究丹参注射液对糖尿病（DM）大鼠心肌纤维化的影响。方法除正常对照组外，采用高脂高糖饲料联合腹腔注射链脲佐菌素（STZ）的方法建立DM大鼠模型。造模成功后随机分为模型组，丹参低、中、高剂量（0．75、1．5、3ml／kg）组。药物干预8周后，生化检测空腹血糖（FBG）、甘油三酯（TG）、总胆固醇（TC）；Masson染色法观察心肌组织形态学变化；采用Western blot技术检测心肌组织p38MAPK、p-p38MAPK蛋白的表达。结果与正常对照组比较，模型纽大鼠FBG、TG、TC升高（P〈0．05），心肌组织胶原表达量增加，心肌间质出现明显纤维化，p38MAPK、p-p38MAPK蛋白表达增高（P〈0．01）。与模型组比较，丹参注射液治疗组的FBG、TG、TC明显降低（P〈0．05），心肌间质胶原纤维明显减少，p38MAPK、p-p38MAPK蛋白表达降低（P〈0．05）。结论丹参注射液能调节DM模型大鼠心肌p38MAPK蛋白表达水平，并可能从而改善DM大鼠心肌纤维化。
Objective To study the effect of salvia miltiorrhiza injection on myocardial fibrosis in diabetic rats （DM） based on p38 mitogen activated protein kinase （p38MAPK） signaling pathway. Methods In addition to the normal control group, a high-fat, high-sugar diet combined with intraperitoneal injection of streptozotocin （STZ） was used to establish a DM rat model. After successful modeling, they were randomly divided into model group and low, medium, and high doses of salvia miltiorrhiza （0.75, 1.5, and 3 ml/kg ） groups. Eight weeks after drug intervention, fasting blood glucose （ FBG ） , triglyceride （ TG ） and total cholesterol （ TC ） were detected, biochemically myocardial morphological changes were observed by Masson staining, the expression of p38MAPK and p-p38MAPK protein in myocardial tissue were detected by Western blot. Results Compared with the normal control group, the FBG, TG, and TC in the model group were increased （P〈0.05） , the expression of collagen in the myocardial tissue was increased, the fibrosis was evident in the myocardial interstitium, and the expression of p38MAPK and p-p38MAPK protein was increased （ P〈0.01 ） . Compared with the model group, FBG, TG, TC （ P〈0.05 ） , myocardial interstitial collagen fibers were significantly decreased, and the expression of p38MAPK and p-p38MAPK protein was decreased in the salvia miltiorrhiza injection group （P〈0.05） . Conclusion Salvia miltiorrhiza injection can regulate the expression ofp38MAPK protein in DM rats and may improve myocardial fibrosis in DM rats.
