摘要
利用高速逆流色谱技术(HSCCC)从荷叶中分离制备高纯度的黄酮类物质,并对所制备的荷叶黄酮进行抗氧化活性研究。结果表明,以正己烷∶乙酸乙酯∶甲醇∶水=1∶6∶1∶6和正丁醇∶甲醇∶水=5∶1∶4作为HSCCC分离的两相溶剂体系,得到三种目标组分C_1、C_2和C_3的质量为12.4、5.9、2.1 mg,经高效液相分析检测其纯度分别为92.67%、90.43%、87.52%。通过标准品对照实验,得到三种目标组分可能为紫云英苷、异槲皮苷及槲皮素。并对目标组分的抗氧化活性进行研究,得到对羟基自由基的半数清除率(IC_(50))分别为0.94、0.85、0.76 mg/m L,对DPPH自由基的半数清除率(IC_(50))分别为0.39、0.28、0.06 mg/m L,对ABTS^+自由基的半数清除率(IC_(50))分别为0.56、0.47、0.29 mg/m L,三种目标组分均表现出良好的抗氧化活性。
Isolation and prepare high-purity flavonoid from lotus leaves by High-speed counter-current chromatography and research antioxidant activity.Results showed that:The two-phase solvent system composed of N-hexane∶ethyl acetate∶methanol∶water=1∶6∶1∶6 and N-butanol∶methanol∶water=5∶1∶4.The quality of the three target components(C_1,C_2and C_3)was obtained as 12.4,5.9 and 2.1 mg and Check the purity by HPLC was 92.67%,90.43%,87.52%.The three target components were likely to be orientin,isoquercitrin and quercetin by standard control experiment.The three target components showed good antioxidant activity,the half-value clearance(IC_(50))of hydroxyl radical was 0.94,0.85,0.76 mg/m L,the half-value of DPPH free radical scavenging rate(IC_(50))were 0.39,0.28,0.06 mg/m L,the ABTS~+free radical Half of the clearance rate(IC_(50))were0.56,0.47,0.29 mg/m L.
