基于Na_(2)S_(2)O_(4)胁迫下的防风色原酮生理生态作用研究 被引量:4

摘要 目的:以O^(-)_(2)的载体Na_(2)S_(2)O_(4)模拟植物环境胁迫,通过不同时间防风体内抗氧化酶活性、H_(2)O_(2)含量及色原酮含量的变化,阐明防风次生代谢产物色原酮在抵御环境胁迫过程中的生理生态作用。方法:研究外源Na_(2)S_(2)O_(4)对防风鲜根体内抗氧化酶活性、PAL活性、H_(2)O_(2)含量及色原酮含量的影响,阐明药材质量形成机理。结果:在Na_(2)S_(2)O_(4)胁迫下防风SOD、CAT、APX活性变化不大,POD活性显著升高;Na_(2)S_(2)O_(4)使防风根内H_(2)O_(2)含量下降,15μmol/L的Na_(2)S_(2)O_(4)胁迫9天后H_(2)O_(2)含量下降幅度最大,为对照组的68.3%;Na_(2)S_(2)O_(4)胁迫后,3、5、7、9、12天防风色原酮含量显著提高,总色原酮含量分别提高14.17%、27.26%、19.87%、24.38%、21.94%;DPPH清除试验显示色原酮本身没有清除活性氧的能力,但与POD同时存在时显著提高H_(2)O_(2)的清除能力。结论:色原酮是防风抵御环境胁迫的主要物质,环境胁迫下色原酮与POD共同作用参与ROS的清除过程。 Objective:To simulate plant environmental stress with superoxide free radical carrier Na_(2)S_(2)O_(4),to study the ecophysiological effects of chromone,a secondary metabolite in Saposhnikovia divaricata roots,on the resistance to environmental stress through the changes of antioxidant enzyme activity,H_(2)O_(2)content and chromone content in Saposhnikovia divaricata roots at different time.Methods:The effects of exogenous Na_(2)S_(2)O_(4)on the antioxidant enzyme activity,PAL activity,H_(2)O_(2)content and chromone content in the fresh roots of Saposhnikovia divaricata were studied,and the quality formation mechanism was elucidated.Results:Under Na_(2)S_(2)O_(4)stress,the activities of SOD,CAT and APX did not change much,but POD activity increased significantly.Na_(2)S_(2)O_(4)reduced the content of H_(2)O_(2)in the roots of Saposhnikovia divaricata.After 9 days of Na_(2)S_(2)O_(4)(15μmol/L)stress,the content of H_(2)O_(2)decreased the most,which was 68.3%of the control group.After Na_(2)S_(2)O_(4)stress,the content of chromone in Saposhnikovia divaricata roots was significantly increased at 3,5,7,9 and 12 days,and the total content of chromone was increased by 14.17%,27.26%,19.87%,24.38%and 21.94%,respectively.DPPH scavenging assay showed that chromone had no scavenging ability of reactive oxygen species,but the simultaneous presence of chromone and POD significantly increased the scavenging ability of H_(2)O_(2).Conclusion:Chromone in Saposhnikovia divaricata roots is the main substance in resistance to environmental stress.Chromone and POD are involved in ROS removal process under environmental stress.
