富血小板血浆对兔膝骨关节炎模型基质金属蛋白酶-9的影响 被引量:1

The Effect of Platelet-Rich Plasma on Matrix Metalloproteinase-9 of Rabbit Knee Osteoarthritis Model

摘要 目的:探讨富血小板血浆对兔膝骨关节炎关节基质金属蛋白酶-9表达的影响。方法:选取36只新西兰大白兔随机分为对照组、模型组和富血小板血浆组(PRP组),每组12只。PRP组、模型组行左膝关节改良Hulth法复制骨关节炎模型,对照组仅打开左膝关节腔。PRP组于造模后第1周取耳静脉血按Aghaloo法制备富血小板血浆,0.5 ml富血小板血浆关节腔注射;对照组、模型组关节腔内注射等量生理盐水造模后第5周,一周注射一次。连续注射6周后处死实验动物,对关节软骨及滑膜组织行免疫组织化学检测基质金属蛋白酶-9的表达量,Western-blot检测软骨细胞中基质金属蛋白酶-9的含量,ELASA检测关节液与血清中基质金属蛋白酶-9的含量,并进行相关性分析。结果:与模型组比较,富血小板血浆可降低关节软骨及滑膜组织中基质金属蛋白酶-9的表达量(P<0.01),但PRP组关节软骨及滑膜组织中基质金属蛋白酶-9的表达量高于对照组,差异有统计学意义(P<0.05);PRP组关节液、血清基质金属蛋白酶-9的含量高于对照组(P<0.05),但明显低于模型组,差异有显著统计学意义(P<0.01)。结论:富血小板血浆可降低骨性关节炎模型中基质金属蛋白酶-9的表达,发挥保护关节软骨的作用,从而延缓骨关节炎的进展。 Objective To investigate the effect of platelet-rich plasma on the expression of Matrix metalloproteinase-9 in Rabbit knee osteoarthritis joints.Method Thirty-six New Zealand white rabbits were randomly divided into control group,model group and platelet-rich plasma group(PRP group),with 12 rats in each group.In the PRP group and the model group,the left knee joint modified Hulth method was used to replicate the osteoarthritis model.The control group only opened the left knee joint cavity.The PRP group took the ear vein blood at the first week after modeling.Platelet-rich plasma was prepared according to the Aghaloo method.0.5 ml of platelet-rich plasma for intra-articular injection;In the control group and the model group,the same amount of saline was injected into the joint cavity in the fifth week,and the injection was performed once a week.The experimental animals were sacrificed after 6 weeks of continuous injection,and the expression of Matrix metalloproteinase-9 was detected by immunohistochemistry on articular cartilage and synovial tissue.The content of Matrix metalloproteinase-9 in cartilage cells was detected by Western-blot,and the content of Matrix metalloproteinase-9 in the joint fluid and serum was detected by ELASA,and the correlation analysis was carried out.Results Compared with the model group,platelet-rich plasma reduced the expression of Matrix metalloproteinase-9 in articular cartilage and synovial tissue(P〈0.01),however,the expression of Matrix metalloproteinase-9 in articular cartilage and synovial tissue of PRP group was higher than that of the control group(P〈0.05).The content of Matrix metalloproteinase-9 in the synovial fluid and serum of PRP group was higher than that of the control group(P〈0.05),but significantly lower than the model group(P〈0.01).Conclusion Platelet-rich plasma can reduce the expression of Matrix metalloproteinase-9 in the model of osteoarthritis,play a role in protecting articular cartilage,and thus delay the progress of osteoarthritis.
