硫化氢对长期过量饮酒小鼠心肌纤维化及miR-221表达的影响

摘要 目的研究硫化氢(H2S)对长期过量饮酒小鼠心肌纤维化的作用。方法 44只实验小鼠随机分成4组:对照组、模型组、硫氢化钠(Na HS)组和DL-炔丙基甘氨酸(PAG)组。除对照组外,其他小鼠采用每日自由饮用4%(Vol/Vol)的乙醇水溶液12 w诱导酒精性心肌纤维化模型。另外,Na HS组小鼠予每日腹腔注射Na HS溶液(50μmol/kg)。PAG组小鼠予每日腹腔注射PAG溶液(40 mg/kg)。12 w后,用VG染色观察胶原沉积,免疫组化测定心肌中Ⅰ型胶原蛋白的表达水平,实时荧光定量PCR(RTq PCR)检测mi R-221的表达水平。结果模型组小鼠心肌胶原沉积较对照组明显加重,Ⅰ型胶原蛋白表达明显增多,mi R-221表达也显著上调(P<0.05)。Na HS组上述指标变化均得到明显改善。相反,PAG组上述指标变化进一步明显加重。结论 H2S具有改善长期过量饮酒导致的心肌纤维化的作用,其机制可能与下调mi R-221的表达有关。 Objective To investigate the effect and potential mechanism of hydrogen sulfide( H2 S) on myocardial fibrosis in chronic excessive alcohol-intake mice. Methods Forty-four mice were randomly divided into control, model, Naris and PAG groups. Except control group, the mice were fed with 4% alcohol for 12 weeks to induce the model of alcoholic myocardial fibrosis. At the same time, the mice in NariS group were intraperitoneally injected with NariS daily at a dose of 50 ;μmol/kg. The mice in PAG group were intraperitoneally injected with PAG daily at a dose of 40 mg/kg. 12 weeks later, collagen deposition was assessed by VG staining. The expression level of collagen I was evaluated by immunohistochemistry, and the expression of miR-211 was determined by real-time quantitative PCR. Results The myocardial collagen deposition in model group was more noticeable than that of control group. Also, the expression of collagen | was significantly increased in model group than that of control group. Meanwhile, the expression of miR-221 was significantly upregulated in model group than that of control group(P〈0.05). All the above changes were obviously relieved in NariS group than those of model group. In contrast, all the above changes were further significantly aggravated in PAG group than those of model group. Conclusions H2S has the effect of alleviating myocardial fibrosis induced by chronic excessive alcohol intake, and its mechanism might be related to the down-regulation of miR-211 expression.
