摘要
目的了解健康人群和丙型肝炎病毒(HCV)-RNA阳性患者血清标本中病毒群落的组成;比较健康组与HCV感染组血清标本中病毒群落组的差异性;挖掘新型HCV及其他新病毒。方法提取血浆样品中的全部核酸,反转录后采用Illumina index primers进行扩增构建病毒文库,随后进行Miseq深度测序,通过生物信息学分析筛选出与病毒相关的序列。结果 HCV01文库中总病毒序列数为1 514条,主要由黄病毒科(48%)、指环病毒科(29%)、反转录病毒科(7%)、微小噬菌体科(2%)构成;HCV02文库中总病毒序列数为2 820条,主要由指环病毒科(61%)、黄病毒科(18%)、反转录病毒科(8%)、微小噬菌体科(1%)构成;HCV03文库中总病毒序列数为3 534条,主要由指环病毒科(78%)、黄病毒科(4%)、反转录病毒科(4%)、人类内源性反转录病毒科(2%)、微小噬菌体科(1%)构成;HCV04文库中总病毒序列数为3 549条,主要由指环病毒科(90%)、反转录病毒科(2%)构成。结论血液中存在多种病毒,HCV感染患者血液中主要存在指环病毒科、黄病毒科、反转录病毒科病毒,而健康人群血液中主要为指环病毒科、反转录病毒科病毒;血清中HCV水平可能对指环病毒科病毒的复制有抑制作用,即HCV水平越高则指环病毒科病毒水平越低。
Objective To understand the composition of virus community in serum sample of both healthy people and patients infected with HCV,to compare the difference of composition of serum sample of health group and patient group,and to discover new genotype hepatitis C virus and new virus.Methods All the nucleic acids in the plasma samples were extracted and the virus library was amplified by Illumina index primers after reverse transcription.Then the Miseq sequence was sequenced and the virus related sequences were screened by bioinformatics analysis.Results The total number of virus sequences in the HCV01 library was1 514,including Flaviviridae(48%),Anelloviridae(29%),Retrovirus(7%),Microviridae(2%);the total number of virus sequences in the HCV02 library was 2 820,including Anelloviridae(61%),Flaviviridae(18%),Retrovirus(8%),Microviridae(1%);the total number of virus sequences in the HCV03 library was3 534,including Anelloviridae(78%),Flaviviridae(4%),Retroviridae(4%),Microviridae(1%);the total number of virus sequences in the HCV04 library was 3 549,including Anelloviridae(90%),Flaviviridae(4%),Retroviridae(2%).Conclusion There are many kinds of virus in the blood,in the serum of patients infected with HCV Anelloviridae,Flaviviridae and Retroviridae are the main viruses,but Anelloviridae and Retroviridae are the main viruses in the healthy group.The level of HCV in serum may inhibit the replication of Anelloviridae.The higher the level of the virus,the lower the level of Anelloviridae.
