摘要
目的:采用响应面法优化黄芪茎叶总黄酮的提取条件,并对其抗炎功效进行评价。方法:在单因素试验的基础上,通过Box-Behnken试验设计,获得总黄酮的最佳提取工艺,并采用小鼠巨噬细胞构建体外炎症模型研究其抗炎活性。结果:黄芪茎叶总黄酮最佳提取条件为乙醇浓度80%、液料比15∶1、提取温度70℃、超声功率200 W,在此条件下总黄酮得率为8.58%;黄芪茎叶总黄酮能够显著降低小鼠巨噬细胞本底NO水平,并抑制由LPS所诱导的NO释放量的增加。结论:利用响应面法分析结果可靠,所得提取工艺合理可行,且黄芪茎叶总黄酮具有明显的抗炎活性。
Objective: To explore the optimal extraction conditions of total flavonoids from stems and leaves of Astragalus membranaceus by response surface methodology and evaluate the anti-inflammatory activity. Methods: Based on the single-factor tests,extraction conditions of total flavonoids were optimized by Box-Behnken design,and the anti-inflammatory activity was assessed by in vitro inflammatory model of mouse macrophages. Results: The optimal extraction conditions of total flavonoids were ethanol concentration of 80%,liquid-material ratio of 15:1,extraction temperature of 62 ℃ and ultrasonic power of 200 W,the maximum extraction rate of total flavonoids was 8. 58% under the conditions. Total flavonoids from stems and leaves of A. membranaceus reduced background NO level of mouse macrophages significantly,and inhibited the production of NO induced by LPS. Conclusion: The results of response surface methodology are reliable and extraction conditions are applicable. The total flavonoids from stems and leaves of A. membranaceus have strong anti-inflammatory activity.
