丁苯酞对慢性酒精中毒大鼠海马线粒体及学习记忆能力的影响 被引量:6

Effects of n-butylphthalide on mitochondria in hippocampus and learning and memory abilities in rats with chronic alcoholism

摘要 目的探讨丁苯酞(n—butylphthalide,NBP)对慢性酒精中毒大鼠海马线粒体和学习记忆能力的影响。方法60只SD雄性大鼠随机均分为正常对照组、模型组和治疗组,每组20只。模型组和治疗组大鼠连续给予6%(V/V)酒精溶液28d,建立慢件酒精中毒大鼠模型,治疗组大鼠在给予酒精溶液第14天开始给予丁苯酞灌胃,灌胃14d。采用Y型电迷宫测试大鼠的学习记忆能力,分光光度法测海马组织H2S的含量和线粒体ATP酶活性,Westernblot检测海马F—actin的蛋白表达量。结果与正常对照组大鼠比较,模型组大鼠学习记忆能力下降,海马组织H2S含量升高,线粒体ATP酶活性和F—actin蛋白表达均降低,电镜下大部分线粒体有损伤。经丁苯酞治疗后,治疗组大鼠的训练次数[(61.88±3.61)次]较模型组[(82.19±4.87)次]降低,学习记忆能力提高(P〈0.05),治疗组大鼠海马线粒体ATP酶活性[(1.84±0.11)U/mgprot]和F—actin蛋白(0.12±0.01]表达较模型组[(1.50±0.07)U/mgprot,(0.08±0.01)]均升高,差异有统计学意义(均P〈0.05)。治疗组大鼠海马组织H2S含量[(34.56±2.47)nmol/g]较模型组[(44.55±3.71)nmol/g]降低,差异有统计学意义(P〈0.05),电镜下线粒体损伤有所改善。结论NBP能改善慢性酒精中毒大鼠的线粒体损伤和学习记忆能力。 Objective To explore the effects of n-butylphthalide (NBP) on mitoehondria in hippocampus and learning and memory abilities in rats with chronic alcoholism. Methods 60 male SD rats were randomly divided into three groups on average, including normal group, model group and treatment group, with 20 rats in each group. Rats of model group and treatment group are given 6% (V/V) alcohol solution continuously for 28 d to establish the model of chronic alcoholism. Rats in the treatment group were given butylphthalide for 14 days from the fourteenth day after giving alcohol solution. The Y type electric maze was used to test the learning and memory ability of rats ,the content of H2S in the hippoeampus and the activity of. mitoehondrial ATP enzyme were measured by spectrophotometry, and the protein expression of F-aetin was detected by Western blot. Results Compared with the normal group, the learning and memory ability of the rats in the model group were decreased, the content of H2S in the hippoeampus were increased, and the activity of mitoehondrial ATP enzyme and the expression of F-aetin protein were decreased,and most of the mitochondria were damaged under the electron microscope.The training times of the rats in treatment group( 61.88 ±3.61 )was lower than that of the model group (82.19.± 4.87), the ability of learning and memory was improved(P〈0.05). Compared with the model group ( (1.50±0.07) U/mgprot, (0.08±0.01) ), the activity of the mitoehondrial ATP enzyme((1.84±0.11) U/mgprot) and the level of F-aetin protein( 0.12±0.01 )in rat hippocampus of treatment group were increased,the difference was statistically significant( both P〈0.05).The level of H2S in rat hippoeampus of the treatment group ((34.56±2.47) nmol/g) was lower than that of the model group ( ( 44.55±3.71 ) nmol/g) , the difference was statistically significant (P〈 0.05 ).
