摘要
目的初步筛选微滤与超滤联用精制淫羊藿水提液的主要工艺参数。方法以总黄酮透过率、溶液固溶物去除率为指标运用单因素平行试验对微滤膜孔径、GE有机超滤膜的截留分子量进行筛选;采用正交设计,以平均膜通量为评价指标,对膜微滤过程中的料液浓度、操作压力、膜面流速三个主要工艺参数进行优化筛选;并检测药液膜精制前、后的粘度、浊度、电导率及p H等理化性质。结果初步确定以0.05μm无机陶瓷膜微滤,前料液生药浓度控制为0.1g生药/m L,膜前操作压力0.15 Mpa、膜面流速为4.0 m3/h,微滤液再进一步用截留分子量为800 Da的GE有机膜进行超滤。结论膜微滤-超滤联用技术对淫羊藿水提液具有良好的精制纯化效果,可有效去除杂质,保留有效成分,改善溶液理化性质。
Objective To preliminary screen the main process parameters of the combined technology withmicrofiltration and ultrafiltration to refine the aqueous extraction of Epimedium brevicornu Maxim. Methods The totalflavonoids transmissivity and solids removal rate were used as indicators, and the single-factor parallel test was used toscreen the pore size of the microfiltration membrane and the molecular weight cutoff of the GE organic ultrafiltrationmembrane. With the average membrane flux as evaluation index, the three main process parameters including concentrationof feed liquid, operating pressure and membrane surface flow rate were optimized and screened by the orthogonal designmethod. The physicochemical properties of solution contained viscosity, turbidity, conductivity and pH were determinedbefore and after refined. Results The inorganic ceramic membrane microfiltration process parameters were as follows: thepore size was 0.05 μm, the raw liquid concentration was 0.1 g crude drug/mL, membrane operating pressure was 0.15 Mpa,membrane surface flow rate was 4.0m3/h. The microfiltration solution was further filtered by GE ultrafiltration membranewith 800 Da. Conclusion The combination of membrane microfiltration and ultrafiltration has a good purification effect onaqueous extraction of Epimedium brevicornu Maxim, which can effectively remove impurities, retain effective componentsand improve the physical and chemical properties of the solution.
