摘要
以白芸豆为原料,采用超声波辅助进行蛋白质(α-AI粗提液)的提取及工艺优化。通过单因素和正交实验,确定了最佳工艺条件为料液比1:10、超声功率150 W、提取时间100 min、提取温度55℃,该条件下蛋白质提取率可达65.93%。在此基础上,将α-AI粗提液进行有机膜分离纯化,得到5种不同分子质量组分Z_(1)(>300 ku)、Z_(2)(100~300 ku)、Z_(3)(50~100 ku)、Z_(4)(10~50 ku)和Z_(5)(<10 ku),对不同组分的蛋白质含量、质量比例、提取率和α-淀粉酶抑制活性进行测定分析。结果表明:蛋白质含量高低依次是Z_(4)>Z_(3)>Z_(2)>Z_(1)>Z_(5),对应组分的质量比例分别为20.98%、17.73%、10.64%、7.12%和43.53%;在一定浓度范围内,α-淀粉酶抑制率与浓度呈线性正相关,α-淀粉酶抑制活性高低依次是Z_(4)>Z_(3)>粗提液>Z_(5)>Z_(2)>Z_(1),对应组分的半抑制浓度值(IC_(50))分别为0.57、1.35、2.21、3.44、4.31 mg/mL和6.09 mg/mL。其中:Z_(4)组分的提取率可达5.48%,IC_(50)仅为0.57 mg/mL,具有较高的提取率和较强的α-淀粉酶抑制活性,达到较好的分离纯化效果,说明超声波联合膜分离是一种较佳的α-AI生产方法。
With white kidney beans as raw materials, ultrasonic assisted extraction of proteins(α-AI crude extracts) was carried out and its process was optimized. Single factor and orthogonal experiments were carried out to determine the optimal conditions which were the solid to liquid ratio 1:10, the ultrasonic power 150 W, the extraction time 100 min, and the extraction temperature 55 ℃. Under these conditions,the protein extraction yield could reach 65.93%. On this basis, the α-AI crude extracts were separated and purified by organic membranes to obtain five components with different molecular weights, namely Z_(1)(>300 ku), Z_(2)(100~300 ku), Z_(3)(50~100 ku), Z_(4)(10~50 ku) and Z_(5)(<10 ku). The protein content, mass proportion, extraction rate and α-amylase inhibitory activity of different components were determined and analyzed. The results showed that the protein content was in the order of Z_(4)>Z_(3)>Z_(2)>Z_(1)>Z_(5), and the corresponding mass proportions were 20.98%, 17.73%, 10.64%, 7.12% and 43.53%, respectively. Within a certain concentration range, the α-amylase inhibitory rate was linearly positively correlated with the concentration. The α-amylase inhibitory activity was in the order of: Z_(4)>Z_(3)>crude extract>Z_(5)>Z_(2)>Z_(1), and the 50% inhibiting concentration(IC_(50)) values of the corresponding components were 0.57, 1.35, 2.21,3.44, 4.31, 6.09 mg/mL, respectively. Among them, the extraction rate of Z_(4)component was 5.48%, while the IC_(50)was only 0.57 mg/mL, suggesting that it had a high extraction rate and better α-amylase inhibition activity, and obtained a good separation and purification effect. Ultrasonic combined with membrane separation is a better method for α-AI production.
